Chinese mitten crab(Eriocheir sinensis)plays an important role in aquaculture industry of China which with high economic value.With the continuous expansion of the breeding scale,the aggressive behavior among individuals in the high-density culture process seriously affects the survival rate and yield,and reduces the economic benefits.Therefore,it is of great significance to explore the occurrence and influencing factors of aggression in each period.In this experiment,we used the continuous camera observation method to quantitatively analyze the aggression behavior and mating behavior,and transcriptome analysis of the genes related to the aggressive behavior,so as to explore genetic factors,fighting experience and physical and chemical signals on its aggressive behavior and the pattern of secondary mating behavior.The results are as follows:The aggressive behavior of E.sinensis is different among different families.Crabs of intrafamily association performed more aggressive acts of shorter duration than interfamily,family B was more aggressive than those from families A and C(P<0.05): family C was the least aggressive,which is a new strain with red carapace.During the second fighting,the intensity and number of fights are significantly different to first fighting and also differed among families,There is a significant difference in family A(P<0.05).The results show that aggressive behavior is related to genetic factors.During the mating period,the two male crabs compete extremely for female spouses with the visual,chemical and tactile sensation.the intensity and number of fights are significantly higher than the existence of a single signal(P<0.05).There is no significant difference between the visual cue and chemistry cue only.The results showed that the source signal of females in mating period had a regulatory effect on male aggressive behavior.Through the continuous camera observation of the behavior of the second mating of male and female crabs of E.sinensis,the basic characteristics of mating behavior were found,and the males had secondary mating behavior,while females did not appear a secondary mating behavior,and the male will not adopt a forced mating strategies.The crab in the resting state was selected as the control group(A).The winner(B)and the loser(C)who had undergone a fierce fight were as the experimental group,and the eye stalk was cut for transcriptome sequencing analysis.The sequencing results showed that the number of differential genes was 432 of A and B,and the number of significant genes was 12;the control group and the loser group are 545 differential genes,and the number of significant genes was 10;the winners and losers screened 536 differential genes,The number of significantly differential genes is 6.Among them,Go classification is annotated to biologicalprocesses,molecular functions and cellular components.KEGG metabolic pathway analysis shows that phage,lysosomal pathway,signal transduction pathway,sugar production and decomposition,and energy metabolism are significantly enriched.This experiment preliminarily explored the effects of genetic factors,fighting experience and physical and chemical signals on the aggressive behavior of E.sinensis,and the mating behavior mechanism of E.sinensis,providing new research ideas for high-density breeding and theoretical basis for genetic breeding of E.sinensis.
