Eucommia ulmoides Oliv.is the Eucommiaceae,endemic to China,with a high value used in industry,herbal medicines,landscaping,et al.With further research of Eucommia resources,the work of hybridization breeding on Eucommia has attracted increasing intention.In this study,genetic variation and genetic parameters of the hybrid progenies were analyzed according to the determination of the growth and economic traits in the third and forth year,which based on to select excellent families and superior individuals,to provide material for the subsequent breeding work.The main results as follows:1.A convenient high performance liquid chromatographic(HPLC)method has been found for the determination of chlorogenic acid and rutin in the leaves of Eucommia ulmoides.Two components was separated by a Aglient ZORBAX SB-C18(4.6mm×250mm,5?m)with a mobile phase of methanol-acetic acid-water(50:1:49,V/V)at a flow rate of 1 mL/min,detection wavelength of 240 nm and column temperature of 32?.2.The height and ground line diameter of hybrid progenies in the third year had a very significant difference between families,and also the leaf-related traits and leaves gutta percha content,but the branch-related traits,chlorogenic acid and rutin content of leaves not.Female parent Xiao Ye had high GCA on the traits of tree height,ground line diameter and secondary metabolites.And male parent QinZhong NO.1 also had high GCA on the traits of tree height and ground line diameter.Hybrids,DaGuo X LongGuai,had a high SCA on the traits of tree height and ground line diameter.Hybrids,HuaZhong NO.2 X ZiYe,had a high SCA on leaves gutta percha content.Hybrids,Xiao Ye X QinZhong NO.1,had a high SCA on leaves chlorogenic acid content.Hybrids,HuaZhong NO.2 X QinZhong NO.1,had a high SCA on leaves rutin content.3.Female parent Xiao Ye had high GCA on the traits of tree height,ground line diameter and leaves gutta percha content.Male parent QinZhong NO.1 had a high GCA on the ground line diameter,chlorogenic acid and rutin content of leaves.The results of two consecutive years had shown that the female parent Xiao Ye and male parent QinZhong NO.1 had high breeding values on the traits of tree height,ground line diameter and the content of secondary metabolites.Hybrids,HuaZhong N0.2×QinZhong NO.1 had high SCA on the traits of tree height and ground line diameter.Hybrids,Xiao Ye×QinZhong NO.1,had a high SCA on leaves gutta content.Hybrids,YanCi X QinZhong NO.1,had a high SCA on the traits of chlorogenic acid and rutin content of leaves.The results of two consecutive years had shown that the female parent YanCi and male parent LuoChao N0.3 had a high GCA on the traits of tree height,chlorogenic acid and rutin content of leaves.Hybrids,Xiao Ye × QinZhong N0.2,DaGuo×LongGuai and DaYe×ZiYe,had a high SCA on the traits of tree height and ground line diameter.Hybrids,Xiao Ye×LuoChao NO.3,had a high SCA on the gutta content.Hybrids,HuaZhong NO.2 X LongGua,had a high SCA on the traits of leaves chlorogenic acid and rutin content.4.Based on the six traits of tree height,ground line diameter,leaf yield,leaves gutta content,chlorogenic acid content and rutin content,three families,NO.2,NO.3 and NO.1,were chosen as the comprehensive utilization families,each traits gained 10.76%,11.27%,-12.13%,9.02%,42.07%and 14.29%compared to the trees planted in experimental zone.Based on the analysis results of tree height and ground line diameter in the forth year the trees planted both in the experimental zone and centralized planting zone,41 superior individuals were selected,and the gains in tree height and ground line diameter were 37.77%and 70.56%.Then according to the five traits of tree height,ground line diameter,leaves gutta content,chlorogenic acid content and rutin content,20 comprehensive utilization individuals were selected,each traits gained 3.05%,1.27%,11.61%,20.81%and 20.04%compared to the primary groups.5.A system ofEucommia marker-assisted selection was constructed,which was used for identification of superior individuals preliminarily selected.Based the results of high growth and MAS,9 superior individuals,1-19,2-32,2-34,1-45,3-29,4-30,2-20,3-55 and 2-23,were selected.The average height and ground line diameter were 4.24m and 36.36mm,each gained 4.52%and 5.80%.
