The PWD(pine wilt disease)is one of the most important and serious diseases attacking pine forest in China.It was well proved that Monochamus alternatus was the main vector of this disease,so population suppression of M.alternatus has been becoming an effective strategy to control PWD.At present,application of chemical attractants plays a direct and effective role in monitoring and suppressing population of this pest.In this paper,we focused on electroantennography response(EAG)of M.alternatus on 12 compounds from different resource,and optimal chromatographic conditions of pheromone attractant explored by GC(gas chromatography)technique.Finally,GC-MS(gas chromatography mass spectrometry)technique was adopted to analyze the components of chemical compound extracted from head,breast and abdomen parts of male M.alternatus.The results were supposed to provide a theoretical reference for optimization or redesign of attractant for M.alternatus,and explore how M.alternatus adapts to different habitats.1.EAG response of M.alternatus to compounds from different odor sourceSexual developmental stage,gender,feeding and diurnal rhythm all played significantly effects on EAG responds of M.alternatus adults when stimulated by selected odors.Responds of females remarkably increased with sexual developmental stage.In sexual mature stage,the responds is significant higher from females than from males.The most in 12 selected compounds could have close relation with mating and oviposition of the adults.It was suggested that the compound A,B,C,L and E had potential uses for composing chemical attractant of M.alternatus adults.2 Gas Chromatographic analysis of M.alternatus aggregation pheromoneBased on standard sample test under five different temperature conditions,standard chromatographic isolated conditions were screened and optimized to get better extracts and compounds.The better conditions were as follow:FID detector,30m×320?m×0.25?m the HP-5 capillary column,the injection volume 1 ?l.The carrier gas was a purity higher than 99.99%pure nitrogen with flow rate of 30 ml/min;igniting gas was 99.99%pure hydrogen gas with a flow rate of 30 ml/min;air flow rate of 400 mL/min,split ratio 50:1,the makeup was 25 ml/min.Temperature program retained the initial temperature of 50? for 5min,then increased to 250?with heating speed of 7 ?/min to rapidly.After testing different concentrations of duplicate samples,standard curve was made as y=327.54x-1.427,the correlation coefficient R2 was 0.9999.To some extent,there was a good linear relationship between peak area dimension and concentration of standard samples.This result provided a theoretical reference for the subsequent development of standard samples.3 GC-MS analysis for extracts from males of M.alternatusTo analyze components of extract from males of M.alternatus by GC-MS,insect body was cut into head,thorax and abdomen parts.18,4 and 4 kinds of compounds were detected in head,thorax and abdomen part,individually.Substantial ?-pinene and 2,4-di-t-butylphenol were found at both head,thorax and abdomen part.It was guessed that the ?-pinene in M.alternatus body might be come from feeding of adults,and it could be the synthesis precursor of semiochemicals,or play important roles on regulating intraspecific behavior such as mating and oviposition.
