| Objective Using multiple PCR to detect Pseudomonas plecoglossicida;Vaccination efficiency of surface antigens of Pseudomonas plecoglossicida;Selectting Chinese herbal medicine which has the bacteriostatic action to Pseudomonas plecoglossicida;With appropriate ways to feed Pseudosciaena crocea,detecting the change of immunological factors and disease resistance,to establish the technology about Chinese herbal prevention and control Pseudomonas plecoglossicida.Methods ①Design and synthesis three pairs specific primers to multiple PCR.②In the present study,main surface antigens of P.plecoglossicida including lipopolysaccharide(LPS),outer membrane proteins(OMP),extracellular biofilm polysaccharide(EPS),and formalin-killed cell(FKC)bacterin,were prepared and the fish vaccinated.On post-immunization day 28,serum antibody titers,phagocytic responses of leukocytes,and lysozyme activities of the fish were evaluated.The efficiency of vaccination was tested by artificial challenge via intraperitoneal injection of live bacteria on post-immunization day 28 and 35,respectively.③Using agar diffusion method to process vitro bacteriostasis experiment,selectting Chinese herbal medicine which has the bacteriostatic action to Pseudomonas plecoglossicida.④Preparation Chinese herbal medicine bait to feed Pseudosciaena crocea for non-specific immunity;detectting the activity of SOD,CAT;Detection Chinese herbal medicine of bacteriostatic action in the bodyResults ①After multiple PCR,Pathogenic strains of P.Plecoglossicida appear 3 items Objective gene.②Although significant humoral and innate immune responses were elicited in all vaccination groups,the challenge produced similar poor protection in both tests.③The selected Chinese herbal medicine including Gulla Chinensis,Radix Scutellariae and Codonopsis Pilosulawhich has the bacteriostatic action to Pseudomonas plecoglossicida.④The group of Scutellaria radix,Astragal us membranaceus and Codonopsis Pilosula showed a certain protection after feeding 28 days,Showed the Chinese herbal medicine has certain immune effect of Pseudomonas plecoglossicidaConclusion ①Multiple PCR is feasible,can be used to Pseudomonas plecoglossicida for rapid identification.②All vaccination groups have a certain immune response,showed using vaccination against the fish pathogen P.plecoglossicida is feasible.③Suggests Chinese herbal medicine additive could effectively improve the nonspecific immune function of Pseudosciaena crocea,provides a certain theoretical basis for Chinese herbal medition using in feeding Pseudosciaena crocea. |
