Fujian Province is one of the best provinces in quantity of edible mushroom and cultivation techniques.In recent years,with the increasement of the support of nation on the modernization agriculture,industrial cultivation of edible mushroom will be a a trend in the future.Most of the edible fungi practitioners haven't much special knowledge in Fujian and don't much care about proprietorial rights of varieties of edible mushroom,So some people don't know what is the cultivated varieties in their cultivated process,hindering the development of edible mushroom industry seriously in Fujian Province.To solve these problems,the following parts were focused:1 To solve the problem of too many samples and amount of work,We.developed a method of DNA extraction based on conventional CTAB DNA extraction.This approach can extract DNA fast and conveniently from many samples,and just need two and half hours in all process.we optimize a method of extracting DNA from fruiting bodies and the result showed that under conditions of 4%CTAB extraction buffer;extraction time 50min and 1%CTAB mediation liquid,we can have high-quality DNA from fruiting body.Meanwhile we have tried to extract DNA from fruiting bodies.2 Based on the national standard of edible fungi cultivar identification,ITS-RFLP,SRAP and SCAR were done to increases its accuracy.Finally,We obtained eleven molecular marker primers that used to identify cultival of Lentinula edodes,Simultaneously obtained eleven primers of Auricularia polytricha,eight primers of Pleurotus geesteranus,ten primers of Agrocybe aegerita and nine primers of Agaricus bisporus.ITS-XbaI digestion reaction of Auricularia polytricha showed that the sample have two species,one is Auricurlaria auricula the other is Auricularia polytricha,ITS-Hind ? digestion reaction of Agrocybe aegerita showed that the strains have two species,ITS-Hae? digestion reaction of Agaricus bisporus showed that the strains have five species.3 In order to prove the stability and feasibility of RAPD?IS SR and SRAP,we have extracted DNA from mycelium of Lentinula edodes,Auricularia polytricha,Pleurotus geesteranus,Agrocybe aegerita and Agaricus bisporus in the same time,and stored them at minus twenty degree Celsius,and then we performed PCR experiments in first,fourth,seventh and tenth days with the DNA after stored,showed that the experiment of molecular markers is stability and feasibility.4 With the purpose of giving every stains a right name,standard cultivated varieties from Sanming My cological Institute,Mycological Research Center of Fujian Agriculture and Forestry University and Institute of Edible Fungi of Fujian Academy of Agricultrual Science were compared with the samples to be identified at the same time.Finally we got a ideal result.5 With the analytical method of manual cultivar identification diagram(MCID),we have made CID of the 235 Lentinula edodes strains distinguished the strains,contained 16 classification.made CID of the 92 Auricularia polytricha strains distinguished the strains,contained 7 classification.made CID of the 15Auricularia auricula strains distinguished the strains,contained 5 classification.made CID of the 71 Pleurotus geesteranus strains distinguished the strains,contained 10 classification.made CID of the 76 Agrocybe aegerita strains distinguished the strains,contained 11 classification.made CID of the 47 strains distinguished the strains,contained 121 classification.This result lays the foundation for subsequent variety identification of edible fungi in Fujian Province.
