The purpose of the study is to establish the model of acute chemical liver injury in piglets by intraperitoneal injection of carbon tetrachloride,and then study the influence of Phyllanthus urinaria L Compound(PULC)on growth performance,liver injury recovery,liver antioxidant activity,and liver structure of piglets.A total of thirty-six castrated piglets of 28±1d were randomly divided into six groups,(namely blank control,challenge,DDB control,low PULC,medium PULC,high PULC groups)with 6 replicates per group.The piglets in the blank control group and challenge group were only fed the basal diet,the piglets in the DDB control group were fed the basal diet supplemented with 20mg/kg DDB,whereas the piglets in the PULC low,medium and high groups were fed the basal diet supplemented with 250mg/kg,300mg/kg and 350mg/kg PULC,respectively.All groups were fed with the diets for 28 days.Then the piglets were weighed and intraperitoneally injected once with either 5%peanut oil(only for blank control group)or carbon tetrachloride(1.5ml/kg body weight).24h post injection,all piglets were euthanized for blood and tissue sampling.Blood sample were used to to investigate the biochemical parameters(ALT?AST?ADA?LDH);liver tissue were used to to observe the microscopic structure of the liver and homogenated liver tissue were used to to investigate antioxidant biochemical parameters(CAT?SOD?T-AOC?MDA).The results are as follows:1.Effects of PULC on growth performance of pigletsThe final weight and average weight daily gain of piglets in PULC high group was significantly higher than that in the blank control group(P<0.05).And there were no significantly differences among the PULC low,medium,DDB control group and blank control groups(P>0.05).And there were no significantly differences in daily feed intake among the groups(P>0.05).The results suggested that PULC can promote the growth performance of piglets in high doses,but it can not increase the daily feed intake of piglets.2.Effects of PULC on piglets anti-CCl4 liver injuryThe activities of ALT?AST?ADA?LDH in the challenge group were extremely significantly higher than that in other groups(P<0.01),and the activities of ALT?AST?ADA?LDH in PULC high group were extremely significantly lower than that in other groups(P<0.01).But there were no significantly differences among PULC low,medium,DDB control groups(P>0.05).3.Effects of PULC on piglets liver antioxidant activitiesThe activities of CAT,SOD,T-AOC in the challenge group were significantly(P<0.05)or extremely significantly(P<0.01)lower than that in other groups,and the activities of them were significantly(P<0.05)or extremely significantly(P<0.01)higher in PULC high group than that in other groups,the effect has a dose dependent manner,as in the lower dose,the activities of liver antioxidant parameters were also lower.The activities of MDA in the challenge group were extremely significantly higher than that in other groups(P<0.01),and the activities of them were significantly(P<0.05)or extremely significantly(P<0.01)lower in PULC high group than that in other groups.4.Effects of PULC on piglets liver tissue microscopic structureCarbon tetrachloride could induce the liver injury model in piglets evidenced by liver tissue pathological changes(disorderd hepatocytes arrangement,hepatocytes swelling andnecrosis,inflammatory cell infiltration,hepatocytes nuclear concentration,fragmentation and disappearence)in the challenge group.But in the PULC treated groups,regardless of the dose,the liver lesions were obviously relieved.Conclusion:Incorporation of PULC in the basal diet can significantly reduce the stress,improve the gastrointestinal digestive ability,promote the growth performance,increase the feed conversion rates,enhance the liver antioxidant activities,and alleviate liver damage.The effects are dose-dependent,as higher dose can give to better results.
