Vaccinium uliginosum is a deciduous shrub in the Ericaceae Vaccinium species.The ripe fruit is rich in anthocyanins and flavonoids,and has high nutritional and economic value.In this study,we use thie ripe fruit of Vaccinium uliginosum.as materials for transcriptome sequencing by using high-throughput sequencing technology.And we use design primers by MISA software.And we study the diversity of germplasm resources on 80 Vaccinium uliginosum samples from 10 different wild populations by SSR polymorphism primers that screened out from the whole.The conclusions are as follows:1.The transcriptome data were analyzed.10,264 SSRs loci were detected among these unigenes by MISA.Among them,single nucleotide repeats3135,dinucleotide 5485,trinucleotide 1554,Tetra nucleotide repeats 67,five nucleotide repeats 8,hexanucleotide repeat 15.In dinucleotide,the ratio of AG/CT is highest.In trinucleotide,the ratio of AAG/CTT is the highest.2.210 pairs of primers were designed and synthesized.A total of 25 pairs of primers with high polymorphism were screened out from the samples from 5 different wild populations.The percentage of polymorphism was about 11.9%.16 pairs were selected for genetic diversity research and flurther cluster analysis of 80 samples which from 10 different wild populations.3.16 polymorphic primers pairs amplified 165 polymorphic bands in 80 Vaccinium uligioosum samples,the average size was 164bp to 299bp,10.3 polymorphic bands were detected by each primer.The number of polymorphic bands amplified by primer VUSSR209 was up to 14,the polymorphic bands amplified by primer VUSSR195 were at least 5.The average effective number of alleles(Ne)was 1.2160.The range of Nei's gene diversity index(H)was 0.0862 to 0.2217,the mean value of 0.1473.Shannon's index(I)was 0.1630-0.3426,with an average of 0.2529.The genetic similarity coefficient of in averaged 0.97.4.Among the 10 wild populations,the H population had the lowest polymorphism and the C population is the highest.The genetic relationship between C and E population is the closest among 10 populations.Analysis of Vaccinium uliginosum.UPGMA clustering,10 wild populations were clustered into five categories,H and J populations clustered into the same one,and differ from the others.All in all,the genetic diversity of Vaccinium uliginosum.Internal population is low by using SSR markers.The differences between H,J populations and other populations were distinguished,but the differences between other populations and various species have little difference.
