In this study, mature fruit of Vaccinium uliginosum Linn. and white-fruited variety were used as material, The conventional method is applied to determination of nutritional index,10 species flavonoid compounds content were determined by HPLC method, application of high-throughput sequencing technologies for the transcriptome sequencing, comprehensive analysis of test results and conclusions are as follows:1. Compared Vaccinium uliginosum Linn. with white-fruited variety mature fruit nutrition indicators, the content of titratable acid and reduced ascorbic acid is lower in white-fruited varieties, the content of reducing sugar, soluble sugar, polyphenol, flavonoid, anthocyanin is higher in Vaccinium uliginosum.2. HPLC method for determination of flavonoids in 10 compounds, among of the coumaric acid, dihydromyricetin, catechin and epicatechin content, white-fruited variety is higher than Vaccinium uliginosum. Kaempferol, myricetin, quercetin, dihydro quercetin, procyanidin B2, cyanidin-3-glucoside content, Vaccinium uliginosum is higher than white-fruited variety..3. By transcriptome sequencing of the Vaccinium uliginosum and white-fruited variety, acquired 34.62Gb Clean data. Clean data of the samples is better than 5.30Gb, Q30 base ratio is better than 88.73%. Received 89,725 spline unigene after assembly, the length of the above 1 kb have 20,473 spline, functional annotation of differentially expressed genes, received 42837 unigene comments results. The total number of genes differentially expressed are 325, which up-regulated expression genes number are 41, down-regulated expression genes number are 284.4. In the flavonoids biosynthetic pathway, a total of 4 differentially expressed the enzymes, up-regulated expression genes number are 3, down-regulated expression gene number is 1. A total of path related genes are 80, DEGs gene number are 3. Flavonoids path structure genes are 11, which up-regulated expression genes number is 9, down-regulated expression genes number are 2. A total of 7 transcription factor differentially expressed, which up-regulated expression number are 5, down-regulated expression genes number are 2.5. By MADS-box and MYB3 transcription factors coordinating role, lead to down-expression of CHI and LAR, affect white-fruited variety fruit coloring.
