A new fermentation technique of protein feed was reported in this study with papermulberry, which is largely and cheaply provided with enriched nutritions,by single and double species fermentation,in order to improve its quality and enhance its percent conversion.Discarded stem leaf of papermulberry was dried, smashed, balanced divided into groups and fermented with Aspergillus niger or/and Saccharomycete. The media optimization of Aspergillus niger fermentation demonstrated that molasses was the optimal carbon source with single factor test by testing reducing sugar and crude protein and counting its spores. The results of optimum carbon-nitrogen ratio showed that as it was 3:1,sugar content was increased 3.16 times reached to 40.79 mg/g after fermentation at 24~27? for 63.5 hours, whereas as it was 2:1,crude protein content was improved 9.58% with natural pH and without additional water in the fermentation process. The optimum carbon-nitrogen ratio of media was carried by 3:1 and 2:1 in Aspergillus niger and Saccharomycete fermentation. When the ratio of carbon-nitrogen was 3:1,the reducing sugar and crude protein reached to 90.08mg/g and 33.75% respectively. As the ratio of carbon-nitrogen was 2:1,the reducing sugar and crude protein reached to 93.89mg/g and 34.01%.Therefore,2:1 carbon-nitrogen ratio was better in the mixed species fermentation. In order to evaluate the biological effects of the fermented feed and its toxicity, stem leaf of papermulberry and its fermentation feed were mixed with mice grain.Non-fermented feed was prepared with 35% stem leaf of papermulberry and 65% standard rat food.Fermented feed was prepared with 35% fermented stem leaves of papermulberry and 65% standard rat food.this experiment,we mixed the fermented leaves and standard rat food in different proportions. The mice were feeded with dried leaves,Non-fermented feed and Fermented feed, standard rat food as the control. The compared experiment was carried out by weighing the mice. The results demonstrated that the dried leaves were not favorite feed to the mice, and Fermented feed was the best protein feed additive as the favorite food of mice and could gained 16.80 g per mouse in two weeks. There is no health problems as protein feed in our test.It was showed that fermentation of papermulberry to make protein feed is ideal and better than standard rat grain in some aspects.
