The mammalian sperm is the foundation of reproduction,comparative proteomics analysis of sperm in livestock play an important role in revealing spermiogenesis and development,capacitation,cryopreservation and other series biologicalphenomena.Sperm samples were collected from the Anhui White goat and Hu sheep.In this study,TMT labei method and LC-MS/MS method were used.A totle of 3 127 proteins were identified in Hu sheep and Anhui White goat sperm,including 2 642 proteins were quantified.we chose the ratios of proteins quantitative of Anhui White goat/Hu sheep greater than 1.5 were defined as the screening criteria.Among these quantified proteins,thereare 553 differentially expressed proteins,290 proteins were up-regulated in the Anhui White goat sperm,while 263 proteins expression increased in the Hu sheep sperm.The 553 differentially expressed proteins were analyzed by GO annotation,in the cellular components,most of the up-regulated proteins of Anhui White goat sperm were located in the cell,organelle,and membrane;in the biological process,most of identified proteins involved in single-organism process,cellular process,metabolic process,biological regulation and so on;in the molecular function,most of identified proteins involved in binding and catalytic activity.We performed a GO enrichment analysis with different proteins of Anhui White goat and Hu sheep sperm.In the molecular function,we found that up-regulated proteins of Anhui White goat sperm involved in Sulfur compounds binding and Vitamin binding activity;in the biological proecss,most of identified proteins involved in sperm motility,flagellated sperm motility,cellular lipid catabolic process,and microtubule bundle formation.By contrast,in the molecular function,we found that down-regulated proteins of Anhui White goat sperm involved in aldehyde dehydrogenase(NAD)activity,oxidoreductase activity,and thiolester hydrolase activity;in the cellular process,the identified proteins were located in the endopeptidase complex,proteasome complex,peptidase complex and so on;in the biological process,most of identified proteins involved in proteasome-mediated ubiquitin-dependent protein catabolic process,telencephalon development,glutamate metabolic process and so on.KEGG pathway analysis revealed that up-regulated proteins of Anhui White goat sperm mainly involved in fatty acid degradation,valine,leucine andisoleucinedegradation,fatty acid metabolism,PPAR signaling pathway,complement and coagulation cascades;while most of down-regulated proteins involved in proteasome,alanine,aspartate and glutamatemetabolism.HSP90A11,such as ACLY,ACACB and LDHC were considered as central hub nodesinthe protein-protein interaction network.
