With the rapid development of sheep industry in China,how to improve the reproductive performance and intensive production of the sheep have received much more attention.At this stage,the quality of mature sperm is one of the main factors that affect the quality of frozen sperm,artificial insemination efficiency and reproductive rate.The maturation of sperm cells is achieved through complex biological processes such as transcription,translation,post-translation modification and gene regulatory networks.Proteins are the diret executant life functions,therefore,in this study,the Label-free LC-MS/MS approach was employed to investigate the proteome profile of spermatozoa of Hu sheep and Anhui white goat,which are the most common sheep breed and goat breed in Anhui Province.Which contribute to uncover the protein-ptotein networks and discover the key genes or biomarkers in the development of sperm cell.Which may provide novel information to understand their physiological function or diversity at protein level.Our results were listed as follows:1.We found that a total of 1993 proteins were identified in sheep sperm.Proteome profile of sheep sperm.We found that identified proteins,of which 18.1% featured membrane-spanning domains and 10.3% harbored signal peptides.On the basis of GO annotation,We identified protein have 89 were involved in fertilization in biological processes.On the basis of COG note,We identified protein have 183 were involved in translation modification and 116 were involved in energy production and conversion.KEGG pathway analysis revealed that the majority of the pathways metabolic and biosynthesis of antibiotic.2.We found that a total of 1943 proteins were identified in goat sperm.Proteome profile of sheep sperm.We found that identified proteins,of which 18.8% featured membrane-spanning domains and 10.1% harbored signal peptides.On the basis of GO annotation,We identified protein have 85 were involved in fertilization in biological processes.On the basis of COG note,We identified protein have 174 were involved in translation modification and 112 were involved in energy production and conversion.KEGG pathway analysis revealed that the majority of the pathways metabolic and biosynthesis of antibiotic.3.Statistical analysis showed that 265 differential expressed proteins were identified between sheep and goat in sperm,173 proteins were up-regulated in the sheep sperm,while 92 proteins were up-regulated.60 proteins were specifically expressed in sheep and 26 proteins were specifically expressed in goat.The PPI network indicated that several proteins,such as Cystatin,a disintegrin and metalloprotease 2,Seminal vesicles 14 kDa protein,3-ketoacyl-CoA thiolase differential expressed between sheep and goat had more interactions with other proteins in the network.
