Ultra-low temperature freezing technology can save the animal sperm for a long time.But in the process of freezing and thawing, it will seriously affect the spermphysiological function and morphology. In the freezing and thawing process, the spermwill be attacked by oxygen free radicals which will directly change the physical andchemical properties membrane,damage to the mitochondrial activity and affect itsvitality. At the same time, it can make changes in protein expression and function whicheventually led to the unsuccessful fertilization. Melatonin can remove oxygen freeradical and has the antioxidant protection effect. So it can protect the sperm fromoxidative damage.In this experiment,we use ram sperm which is devided into melatonin treated with1mM group and non-melatonin treatde group. Use2-DE constructed of melatonin onram sperm to detect the differences in the two groups after freeze-thaw sperm proteinexpression profiles.Then use PDQuest8.0software to analyze protein points on gel mapand detect differential proteins.The results showed:(1)1mM melatonin treated sperm had a higher motility rate thannon-melatonin treated sperm.We use17cm(pH3~10)IPG strips and the suitable samplequality was0.45mg after the gels stained with Coomassie Brilliant Blue.(2) We foundtwelve differential protein sopts. Five spots were increased and seven spots decreased inmelatonin treated sperm2-DE gels. The spots were analyzed and a search againstprotein sequences in the National Center for Biotechnology Information databasesindicated that the differentially expressed proteins correspond to five proteins.They arearylsulfatase A、chromosome21open reading frame33、mitochondrial superoxidedismutase2、alpha-enolase isoform3and cAMP-dependent protein kinase type.
