Rice(Oryza sativa L.)is one of the most important crops in China.it is also the staple food of 2/3 people in the world.The nutritional value of rice plays an important role in the diet.As a special new rice variety,functional rice has some special ingredients that can regulate and balance the physiological functions of the human body.Anthocyanins are water-soluble natural pigments in plants and belong to a class of flavonoids.They are found only in certain rice varieties(e.g.black rice varieties).Total flavonoids are flavonoids and belong to a wide range of natural products,which are widely found in the vegetable kingdom.Anthocyanin and total flavonoids have the medical function to human body.As the target of rice breeding,the research of anthocyanins and total flavonoids is less reported.Zibaoxiangnuo 1,cultivated by Yushan Special Rice Research Institution,is a special functional rice and contains flavonoids.To study Flavonoids composition and inheritance law of Zibaoxiangnuo 1,this experiment has analyzed the total flavonoids and anthocyanin content of recombinant inbred line population(RILs)derived from Zibaoxiangnuo 1 and Minghui 63,the male sterile lines and restorer lines breeded by Zibaoxiangnuo 1,the main results are as follows:1.The optimum conditions for the determination of anthocyanin in brown rice samples were found by optimizing the conditions of liquid chromatography.We respectively weigh 5.0 mg six kinds of anthocyanin standard,the solution is dissolved in10%hydrochloric acid methanol solution and fixed to 10mL volumetric bottle respectively,and the single standard storage liquid is gradually diluted with 10%hydrochloric acid methanol as 0.5mg/L、1.0mg/L、5.0mg/L、10.0mg/L、25.0mg/L,the analysis showed that the six standard solutions of anthocyanins were linear in the range of 0.5 to 25 mg/L,and the linear equation of anthocyanin is linear y=12786.4x-67811.53,R~2=0.9992;linear equation of paeoniflorin y=130649.59x+14073.25,R~2=0.9998;six kinds of anthocyanin concentration of 10 mg/L HPLC chromatogram separation is good,0.5~2 mg/L concentration range of the rate of recovery was 88.6%~110.4%,the precision of the method(RSD)is 1.2%~4.6%,the detection limit was 0.15 mg/kg.The results showed that the stability of the brown rice was high and the data was relatively accurate.For the liquid phase map,the samples of the brown rice were determined only by the two types of anthocyanins,which contain a combination of Cyanidin and Peonidin.2.The anthocyanin content of RILs was measured continuously for three years.The results showed that different levels of anthocyanins in the RILs population were different,the line with highest content of anthocyanidin is No.1 and the anthocyanidin is1894.75 mg/kg,which is obviously superior to the parents.But due to geographical differences,anthocyanin content of some lines had some difference indifferent years,which indicated that the planting location,temperature,light may have a certain effect on the anthocyanin content.Compared with the content of anthocyanin of the sterile lines and restorer lines breeded by Zibaoxiangnuo 1,the content of anthocyanin of Zibaoxiangnuo 1 and the male sterile lines is less than restorer lines,which indicated that the restorative system of the high anthocyanin content of the Zibaoxiangnuo 1 is very effective.3.The content of anthocyanin in Zibaoxiangnuo 1 was determined at different filling stages.The results showed that the content of anthocyanin showed a S shape curve,and the high value of anthocyanin was 16~24 d,and the anthocyanin content decreased slightly at the ripe stage.Minghui 63 does not contain anthocyanin.4.The content of total flavonoids in RILs samples was determined by spectrophotometry.The results showed that rutin standard solution in 5~100 g/mL has good range of linear,y=268.9x+0.5227,R2=1,5~25 g/mL concentration range in the recovery rate of 92.99%~116.84%,the precision of the method(RSD)is 0.3%~1.5%,the results show that the detection limit was 0.025 g/100g,which means that the 5~100μg/mLis good for the total flavonoids measurement.5.The total flavonoids in Zibaoxiangnuo 1 were 0.16%,and Minghui 63 was 0.15%.for the RILs,the content of total flavonoids were stable in different years,indicating that the content of total flavonoids was less affected by environmental factors,such as planting area,light,temperature and so on.Otherwise,the line with high total flavonoids content is No.25(0.33%),which was obviously better than that of the parents.6.The analyze of the change of total flavonoids in different filling stages for Minghui 63 Zibaoxiangnuo 1,which showed that the content of Zibaoxiangnuo 1 and Minghui 63 showed an opposite S shape curve in grain filling stage,and total flavonoids content 0~24 d had reverse connection,The trend of 24~32 D total flavonoids content decreases and reaches a steady state.
