FAM134B and A-FABP play an important role during the deposition of intramuscular fat(IMF).In order to further explore those genes' expression and association with the goat meat,we use RT-PCR to amplify the cDNA sequence of FAM134B and A-FABP in Tianfu mutton sheep.At the same time,we examined the spatial temporal expression of FAM134B and A-FABP.An association analysis was made between the content of IMF and the two genes' mRNA expression.Results are as follows:1.We obtained the whole sequence of FAM134B and A-FABP.It shown that FAM134B consisted of 1071bp encoding 356 amine acid(aa)resides.Phylogenetic tree based on the gene sequence suggested Tianfu mutton sheep and cow had a relatively closer affinity.A-FABP consisted of 399bp encoding 132 aa,phylogenetic tree shown that Tianfu mutton sheep and sheep had a relatively closer affinity.2.The expression of FAM134B and A-FABP were determined in eight muscular tissue at 90,180 and 270 days by using RT-PCR,the results showed that FAM134B and A-FABP were expressed at all time points in the eight tissues.The expression of FAM134B were increase first,and then decrease in semimembranopsus,triceps muscle of arm,biceps femoris muscle,gluteus medius,quadriceps femoris and longissimus dorsi.FAM134B has the highest relative expression in longissimus dorsi at 9-month age.In the biceps femoris muscle,the mRNA expression of A-FABP showed an age-related increase,and reached their highest expression at 9-month age.3.Through the residual analysis,we determined the IMF in the eight-muscular tissue of Tianfu mutton sheep.Our data showed that IMF increased in the semimembranopsus,gastrocnemius muscle,longissimus dorsi with age.And the IMF content was significant higher in 9-month age than in 3-month age(P<0.01).however,the IMF content was decreased in biceps femoris muscle with age,reach to lowest value at 9-month age.4.The results of the association analysis show that the expression of FAM134B and A-FABP had a significantly positive correlation with the content of IMF in longissimus dorsi(P<0.05,r=0.991),and the expression of A-FABP in biceps femoris muscle was significantly negative associated with the expression of IMF content(P<0.05,r=-0.657).
