Wound healing is the process of repairing one's own tissue after being traumatized.It is to make the original cells proliferate and differentiate into new cells or tissues after injury to replace the damaged cells and tissue structure.The inflammatory response stage,cell proliferation stage and tissue remodeling stage in wound healing complement each other and repair the tissue together.Trauma has always been the most common problem in veterinary clinics.Bite,frustration and other injuries often occur in animal life.Trauma not only affects the quality of life of animals,but also reduces the level of animal production,therefore,accelerating wound healing is a long-term clinical problem in veterinary medicine.SVF is cell precipitation which is extracted from the body's own adipose tissue after digested by collagenase and centrifuged to remove the mature adipocytes.It is a kind of cell group containing many kinds of cells and it has the same potential of proliferation and multi-differentiation as stem cells and can promote tissue regeneration.The purpose of this study was to investigate the healing effects of autogenous SVF on canine skin trauma in dogs,and to explore how SVF affects the healing rate of wound healing.It can provide a method for the treatment of wound healing in veterinary clinical practice in the future.In this experiment,6 Chinese rural dogs were divided into experimental group and control group,8 square wounds of 1.5 cm×1.5 cm were cut from the back of each dog and 4 wounds from each side.One side was defined as the experimental group and the other side was defined as the control group.The fat extracted from the dog's own groin was made into SVF and injected it into the wound of the experimental group.Each wound of the experimental group was injected with0.5ml cell suspension(5×10~4 cells per cm~2 wound)and the control group was injected with0.5ml normal saline.The area of the wound was determined by ImageJ software on the 3rd,7th,14th and 21st days,calculated the healing rate and recorded the wound healing time.The skin samples were cut on the 7th,14th,21st and 28th days for histomorphologic observation and proliferating cell nuclear antigen(PCNA)immunohistochemical experiments to observe the inflammatory response of the cells.The average number of vascular strips and the proliferation of cells were recorded.The content of vascular endothelial growth factor(VEGF),epidermal growth factor(EGF),platelet-derived growth factor(PDGF)and hydroxyproline(HYP)in tissues were determined by kit.The results showed that the healing rate of the experimental group was significantly higher than that of the control group on the 7th and 14th days after trauma(P<0.05).The average healing time was about 22.5±1.38 days in the experimental group and 27.17±0.75 days in the control group.The healing time of the experimental group was also relatively short(P<0.05).The results of pathological section showed that the inflammatory cells in the experimental group were relatively less than those in the control group on the 7th day,and the number of vascular strips was more than that in the control group(P<0.01).On the 14th day,there were a large number of collagen fibers in the experimental group,a few inflammatory cells in the control group and the number of blood vessels in the experimental group was still more than that in the control group(P<0.05).On the21st day,the wounds were completely healed in the experimental group and the skin hierarchy was complete,while there was still a defect in the epidermis of the control group.On the 28th day,there were signs of regeneration of skin appendages and collagen fibers tended to be arranged in an ordered arrangement in the experimental group,while there was no regeneration of the skin appendages in the control group.The results of immunohistochemical showed that the integral optical density(IOD)of PCNA in wounds of the experimental group was significantly higher than that of the control group on the 7th,14th,and 21st days(P<0.01).The results of ELISA showed that the content of VEGF in the wound tissue decreased gradually,and the content of VEGF in the experimental group was higher than that in the control group on the 7th,14th and21st days(P<0.05).The content of PDGF also decreased,the experimental group was significantly higher the control group on the 7th day(P<0.05)and it was higher than the control group on the 14th day(P<0.05).The content of EGF increased gradually and the content in the experimental groups was significantly higher than that in the control group on the 14th,21st and28th days(P<0.01).The content of HYP also increased gradually and the content in the experimental group was significantly higher than that in the control group on the 14th and 21st days(P<0.05).The results showed that SVF can promote the formation of blood vessels in the wounded granulation tissue,increase the level of growth factor,promote cell proliferation and collagen synthesis at the wounded site and accelerate wound healing in the process of wound healing.
