The aim of this study is to provide more SSR markers as a powerful tool for t study of the molecular level of proso millet,and providing some reference for the breeding of proso millet varieties in the future.In this experiment,ninty-six proso millet resources from 15 provinces(autonomous regions)of six ecotopes were selected as the test materials.First of all,it was phenotypic diversity analysis,then,151 SSR markers of four,five and six bases obtained by transcriptome sequencing were screened by 6 proso millet species with different geographical regions.Genetic diversity analysis in 96 proso millet species by using 85 SSR markers which screening out from 151 SSR markers.The main analysis results are as follows:1.The agronomic traits of 96 plots were investigated,and the highest coefficient of variation was the number of tillers is 42.19%,the lowest in leaf length is 18.21%.Shannon index highest is 2.08 in plant height,the lowest in leaf length is 1.78.Through the analysis of the correlation analysis,the principal component analysis and the use of membership function of 96 proso millet varieties of comprehensive analysis found that:The proso millet from the Gansu Province,Yuzhong County,No.18 yuzhong huangmi highest value is 0.738,the best comprehensive evaluation;The lowest value of the yellow proso millet from Xiaoyi,Shanxi Province is 0.231,and its comprehensive evaluation is the lowest.The cluster analysis of 96 proso millet was divided into four categories,proso millet of group one's the plant height was higher,the length of panicle was longer,the number of tiller was moderate,the number of main stem was more,the thousand seed weight was medium,and the leaf size was moderate;Proso millet of group two's proso millet's plant height is moderated,the spike length is longer,the number of tillers is less,the number of main stem nodes is moderated,the thousand seed weight weight is smaller,the leaf is also larger;Proso millet of group three's plant height and panicle length were lower,the number of tillers is moderate,the number of main stem nodes was less,the thousand seed weight was moderated,the leaf was smaller.Proso millet of group four's plant height is moderate and panicle length were lower,the number of tillers is more,the number of main stem nodes is moderate,the thousand seed weight is heavier,the leaf is moderate.2.Based on the transcriptome sequencing results of the proso millet,151 pairs of SSR primers were designed and using six different varieties of proso millet for screening,a total of 85 pairs of primers were screened,with an average PIC of 0.5119,which was all belong to high polymorphism primers,the Rp value is between 1.00-5.75 and the average is 3.153,where RYW42 shows the smallest Rp value of 1 and the highest Rp value is 14.75 for RYW47,so the selected SSR primers have moderate discrimination,Can be used as the next step in the study of effective molecular markers.At the same time,the SSR amplification system and the reaction system were optimized.To lay the foundation for the accurate genetic diversity analysis of the proso millets resources.3.Using the 85 pairs of SSR primers to analyze the genetic diversity of 96 proso millet resources,a total of 232 loci were detected,with an average PIC of 0.4723,the highest of which was RYW8,with the PIC of 0.7281 and a minimum of RYW15 with the PIC of 0.0400.4.Based on UPGMA,96 proso millets from six ecotopes were clustered,the result is that the six proso millet ecotopes were divided into three groups,Group I be classified as a class of the South autumn&winter-sowing ecotope alone.Group II be classified as a class of the North spring-sowing ecotope,northwest spring-sowing ecotope and Loess Plateau spring-sowing ecotope.Group III be classified as a class of northeast spring-sowing ecotope and northern spring-sowing ecotope.Cluster analysis of 96 proso millets was divided into 4 groups,the first group belongs mainly to Loess Plateau spring-sowing ecotope;The second group belongs mainly to northern spring-sowing ecotope;The third group mainly belongs to northeast spring-sowing ecotope;The fourth group mainly belongs to north spring-sowing ecotope.Overall,the main geographical distribution between the various groups is consistent with the basic geographical distribution,which means that the differences between the different proso millet species created the diversity of proso millet varieties.5.There are two obvious inflection points in the genetic structure analysis of the proso millets resource,K=2,divided into two groups,group I is red(60),mainly from northwest spring-sowing ecotope,north spring-sowing ecotope and Loess Plateau spring-sowing ecotope,Group ? is green(36),mainly from northern spring-sowing ecotope.K = 4,divided into four groups,the group I is red(19),mainly from the spring area of Loess Plateau spring-sowing ecotope,the group ? is green(22),mainly from northeast spring-sowing ecotope and northern spring-sowing ecotope,the group ? is blue(20),the mainly from north spring-sowing ecotope and Loess Plateau spring-sowing ecotope,group IV is yellow(35),mainly from Northwest spring-sowing ecotope.The results are similar to cluster analysis,and the distribution is closely related to the region.
