As glyphosate is used as the representative herbicide,the use area is expanding,and the area of the areca etiolation in hainan is spreading,it is thought that the occurrence of areca etiolation in hainan is related to the herbicide residues in areca forest,so that the residual problem of glyphosate in the hot zone soil is of great concern.Bioassay glyphosate residues was established in this experiment,and using HPLC method to verify the results of bioassay method,at the same time,the herbicide residues in the soil samples of Areca forest in different regions were determined by using HPLC method,and the relationship between herbicide residues and the areca etiolation was preliminarily explored.The main contents are as follows:(1)Seven cities and counties in hainan areca cause of etiolation,and the usage of herbicide were investigated:areca etiolation in hainan(in the investigation of 52 areca forests,42 is etiolation).Herbicides are commonly used in areca forests,and the longer the herbicide is used,the more often it is used,the areca grows worse.The main herbicides used in areca forest are glyphosate ammonium salt,isopropyl ammonium salt and soluble powder,glyphosate,dimethyl glyphosate,glyphosate,glyphosate,etc.(2)Glyphosate residues in the soil of the establishment of bioassay method.take corn,amaranth as the object,the inhibition rate of glyphosate on corn root length and root length of amaranth was taken as evaluation index,combined with the regression curve equation analysis,the results show that glyphosate for corn length of taproot and roots of amaranth has inhibition,and glyphosate their inhibition rate and linear correlated to the concentrations of glyphosate,relevant equations respectively:Y=13.108X-3.1106(r= 0.9755,p<0.9755)and Y=12.541X-4.8611(r=0.9883,p<0.01),the correlation is wonderful,also has the very good stability and repeatability,therefore,both sensitive indicator plant residues can be used in the detection of glyphosate.By using HPLC method to verify the results of bioassay method,the test results show that the measured value and liquid chromatographic values close to raw measurement method by using cheap materials accessible,detection method is easy to grasp,the advantages of easier to popularization and application.(3)Preliminary study on the relationship between areca etiolation and herbicide residues in hainan.Areca forests in hainan mainly uses glyphosate herbicide,some glyphosate may also be added with 2,4-D butyl ester to improve the effect.This experiment of eight cities and counties in different soil types of Areca forests soil sample(in addition to the west of haikou,all sample to have the history)using herbicide glyphosate and 2,4-D butyl ester residues determination,the results show that the acquisition of all soil samples were not detected in the residues of glyphosate and 2,4-D butyl ester shows that areca etiolation have no direct relation with the herbicide residues.
