| The miRNA in Exosome have the function of regulating animal growth.miRNA(microRNA)is a type of non-coding small RNA with a length of about 22 nt,which can be used in animal growth by targeting degradation of mRNA or inhibition of protein translation.The development process plays an important role.In order to further investigate the regulation mechanism of miRNA regulation in Exosome on the growth and development of Yanbian yellow cattle,the miRNA were isolated from Exosome of Yanbian yellow cattle and Hanyan cattle.The expression of miRNA in Exosome blood of Yanbian yellow cattle and Hanyan cattle was analyzed by RNASEQ and bioinformatics technology.The test results are as follows:1.The total RNA in blood Exosome of Yanbian Yellow cattle and Hanyan cattle were extracted and their purity and quality were determined.The results show that the total RNA obtained is mainly 5S RNA,indicating that the integrity of the obtained total RNA samples is good.The OD values under 260nm and 280nm were 1.8-2.2,which indicated that the extracted RNA was of good quality and no other impurities,which could be used for further analysis.2.The small RNA in the blood Exosome of Yanbian yellow cattle and Han Yan cattle were sequenced.The results showed that the small RNA library of Yanbian yellow cattle blood Exosome obtained 18551862 original sequences,and the small RNA library of Han Yan cattle blood Exosome obtained 17203718 original sequences.The base error rate is low and the quality of sequencing is high.The original sequence data showed that the proportion of clean reads in the blood Exosome of Yanbian yellow cattle was 93.09%,and the proportion of clean reads in the blood Exosome of Han Yan cattle was 82.74%.The quality of detection was more than 80%,which met the requirement of sequencing3.The nucleotide length of blood Exosome small RNA samples from Yanbian Yellow cattle and Hanyan cattle were statistically analyzed.The results showed that the length of small RNA sequences of Exosome in blood of Yanbian Yellow cattle and Hanyan cattle were mainly distributed in 17-30 nt.4.The results showed that 912 known miRNA and 115 newly discovered miRNA in Yanbian yellow cattle blood Exosome,905 known miRNA and 317 newly discovered miRNA in Han Yan cattle blood Exosome.The new miRNA prediction results showed that 187 new miRNA were detected in two breeds,including 41 new miRNA in Yanbian cattle and 146 new miRNA in Hanyan cattle.5.The results of differential expression analysis showed that there were 524 differentially expressed miRNA in Yanbian Yellow cattle and Hanyan cattle,among which 271 were up-regulated and 253 were down-regulated.There are 382 known miRNA and 142 new miRNA.6.The results of differentially expressed miRNA target genes showed that 52348 target genes were predicted in two breeds of cattle.The results showed that these target genes were enriched to 3473 biological processes,394 cell components and 879 molecular function.These predicted miRNA target genes were annotated into 326 signaling pathways,the most highly enriched of which was the mTOR signal transduction pathway.To sum up,we obtained the differentially expressed miRNA species in the blood Exosome of Han Yan cattle and Yanbian Yellow cattle,and analyzed the target genes of differential expression of miRNA,GO and KEGG.In order to select the key differential expression miRNA to study how the miRNA regulates the growth and development of Yanbian Yellow cattle and Hanyan cattle through the key signal pathway.Therefore,the mechanism of influencing the growth and development difference between Bian Yellow cattle and Han Yan cattle at the molecular level was discussed and the experimental basis was provided. |
