Study On Flowering Characteristics And Optimization Of Tissue Culture System Of Nobile Type Dendrobium

Nobile type Dendrobium is a species of orchidaceae,mostly flowering in spring.Den.Stardust'Firebird',Den.Stardust'Chiyomi',Den.Rainbow Dance'Akane'are three multiflower varieties of nobile type Dendrobium.Its plant type is short,the flower color is gorgeous,has a large amount of flowers,and the blooming time is long,the ornamental value is extremely high.Compared with other nobile type Dendrobium,the three varieties ofnobile type Dendrobium have shorter cultivation time and easy to flower.There is a significant advantage in the variety of potted flowers.At present,the three species of nobile type Dendrobium have low marketization in China.In order to explore the flowering characteristics of nobile type Dendrobium and speed up the propagation speed,the flowering period and flower organs of nobile type Dendrobium were studied in this study,which lay the foundation for the regulation of flowering and the classification of nobile type Dendrobium.The rapid selection of the proliferation and root culture medium of'Fire bird'was made to optimize the fast of'fire bird'.The results of the study are as follows:1.The flowering characteristics of three varieties of'Fire bird','Chiyomi'and'Akane'were studied.The results were as follows:the flowering period of three varieties of nobile type Dendrobium was different,the flowering time of'Akane'was early,and the difference of lip and stamen column of the three varieties of nobile type Dendrobium was obvious.2.The embryo culture of'Fire bird'is carried out to observe the various stages of embryo growth.The results are as follows:the seed germination of'Fire bird'needs 15d time,embryo culture and seedling should pass through the process of embryo,protocorm,seedling and seedling,and the quality of the protocorm formed is important to its later growth.3.Taking the sterile seedlings of'Fire bird'as the research object,we optimized the proliferation medium of aseptic seedlings.The proliferation medium suitable for'Fire bird'is Hyponex 1;the combination of suitable hormone combination is NAA0.2mg.L^-1+6-BA1.0mg.L^-1;the suitable organic additive is coconut juice of 100mg.L^-1,the multiplication coefficient is the highest,and the plant grows well.Suitable proliferation conditions were:Hyponex 1 3.0g.L^-1+NAA0.2mg.L^-1+6-BA1.0mg.L^-1+coconut juice 100ml.L^-1.4.Based on the'Firebird'seedlings as the research object,the aseptic seedling rooting medium were optimized:The rooting medium of'Firebird'is:Hyponex 1 1.5g.L^-1+Hyponex 21.5g.L^-1,the root number of seedlings is 8.46,and the organic additive of nobile type Dendrobium is 100ml.L^-11 coconut juice,the root number is the most and the root rate is the highest.The best root hormone of nobile type Dendrobium is NAA0.5mg.L^-11 root number is more,it is rooting the best choice of hormones.The suitable seedling rooting medium:Hyponex1 1.5g.L^-1+Hyponex 2 1.5g.L^-1+NAA0.5mg.L^-1+100ml.L^-1Coconut Juice.
