Studies On Different Plantlet Formation Ways Of In Vitro Propagation Of Nobile-type Dendrobium

Nobile-type Dendrobium---one of the most important orchid plants with high ornamental value is now having a bright market prospect.Tissue culture technique is an important approach to industrialized production of Nobile-type Dendrobium.This experiment established the reproduction system of Nobile-type Dendrobium using D.Yohkou 'Peach boy',D.tomoflake'Akebono',D.snowflake 'Red star'etc.through clustered shoots and protocorm induction seperately,focusing on the systematic research of each growth stages to provide scientific basis for industialized production of Nobile-type Dendrobium.The results showed that,the best startup material for clustered shoots propagation is pseudobulb stem segments with axillary buds and the best startup medium is MS+6- BA 1.0 mg/L +KT 0.5 mg/L +NAA0.1-0.2 mg/L;the optimal multiplication medium is MS+6-BA 2.0 mg/L+IBA 0.2 mg/L +mashed potato 50-100 g/L.Bud tips could be used to induce protocorm and this was never found reported before.The optimal medium is MS+KT 2.0 mg/L+NAA 0.2 mg/L;dark condition or 0.5 -1.0 g/L activated charcoal is advantageous to induction.Protocorm can also be obtained through callus tissue,which is induced by upper stem slices of plantlets on the medium of MS+2,4-D 0.2 mg/L+agar 0.3%.The optimal medium for transformation from callus to protocorm is MS+6-BA 1.0 mg/L +NAA 0.2 mg/L.The optimal multiplication medium for protocorm is MS+TDZ 0.05 mg/L+NAA 0.2 mg/L+LH0.5-1.0 g/L+agar 0.3%;the optimal plantlet differentiation medium is MS+TDZ 0.1 mg/L+IBA 0.2 mg/L.The optimal medium for plantlet rooting and strengthening is 1/2MS+KT 0.2 mg/L +NAA 0.5 mg/L+mashed banana 50-100 g/L+chlormequat 1.0 mg/L+sugerl5 g/L.The optimal transplanting medium is pearlite:water moor 1:1...
