Dendrobium nobile is an important ornamental flowers of orchids, Potted of Dendrobium nobile is novel, and it meets the required different novelty in the consumption characteristics of people. With high technical content and profit margins, potted of Dendrobium nobil has significant advantages in the flower industry. In order to provide theory for solving the tissue culture system of Dendrobium nobil is studied. The major achievements are as follows:(1)The effect of different medium components on proliferation of stem: MS basic medium is better than others on the proliferation of adventitious buds of stem; in the carbon source choice, 3% sucrose is best, glucose and fructose are chsoened as carbon source, either increase or budding rate of biomass is relatively low. Compared with apple juice and banana juice, coconut milk is the best in organic amendments ; Growth regulators to 0.05mg / L of TDZ proliferation is the best.(2)The growing point of tip induced PLB: Getting growing points of tips under the dissecting microscope, induced and proliferated the PLBs of Dendrobium nobile. The most suitable basic medium is MS in the induction of PLB, and the most suitable growth regulator substances is 2,4-D, which concentration is 0.2mg/L. NAA in concentration 1.0mg/L which is best to proliferation of PLBs, and growing is better, and the coefficient of the multiplication of PLBs is 3.55 times, NAA in concentration 0.5mg/L which is best to the class differentiation of PLBs, Respectively, the induction, proliferation and differentiation culture conditions are: MS +6- BA2mg/L + NAA0.5mg/L +2,4-D0.2 mg/L, 1/2MS + KT0.05mg / L + NAA1.0mg / L, 1/2MS + BA0.5mg / L + NAA0.5mg / L.(3) Induced proliferation of clustered shoot in basal: using of the base of stem tissue (basal), the results showed that 1/2MS + Cw150ml/L + KT0.5mg/L + NAA2.0mg/L is best, and the induction rate is 81.4%,and the best concentration on proliferation of clustered shoot is MS + KT0.5mg/L + NAA0.5mg/L .(4) Seedling and roots: Added coconut milk to the medium of MS +0.5 mg/LIBA +0.1 mg/LNAA, it is the interests of rooting and seedling, at the same time added the optimal concentration of activated charcoal is 1g/L. When regulated of plant growth hormone, Dendrobium nobile growing is good in the root of the number, length and thickness uniformity, the medium is MS +1.0 mg / L NAA +1 g / LAC.(5) Transplanting and domestication: unscrew the cap of good rooting plantlets before transplanting, and hardened 5 days in tissue culture outdoor, and then removed the cap and transplanted the vitro-seedlings in 1 day. The results that the average survival rate is 84%; The acclimated seedlings are cultured in Light incubator, and the light intensity is setted 1500Lx, Dendrobium nobile growing well and the survival rate is 85.9%, The acclimated seedlings are cultured in Light incubator, which temperature is setted 25℃, under the conditions the survival rate is 86%, and the seedlings growed new roots more easily, growth trend more robust; Under the air humidity which is 85%, the acclimated seedlings is optimum in growth index, the survival rate is 83.6%.
