The lotus is an aquatic herb,belonging to the Nelumbonaceae family and Nelumbo genera.The flowers are large and beautiful and the leaves are nearly round and have high ornamental value.Lotus also has very high economic value because lotus leaf can be medicine and lotus-root is edible.The flowering phase of single flower of lotus only has a week or so which cannot satisfy long time to admire.This experiment mainly studied the growth and development of lotus leaves and petals after different treatment.The enzyme activity and malondialdehyde contents and expression of related genes of lotus blooming of the flower period of lotus and were measured.The purpose of this experiment is to provide a theoretical basis for the study of the growth of lotus flowers and extension.The main results are as follows:(1)Effects of different concentrations of exogenous hormones on the growth morphological indexes of lotus.The lotus were treated with different exogenous hormones.The effect of treatment of 150mg/LGA3?100mg/L 6-BA?100mg/L IAA on leaf area of lotus leaves was most significant.The treatment of 150mg/LGA3?150mg/L 6-BA make lotus growth which was the largest.High concentration of gibberellin treated the number of lotus and the number of flowers were higher than other treatments.Gibberellin causes the lotus to bloom early and extends the flowering senescence period.High concentrations of auxin and cytokinin prolong the initial flowering period.These treatment lead to extend the effect of the flowering period.(2)Effects of different concentrations of exogenous hormones on the enzyme activity and malondialdehyde contents.According to the study,GA3 and 6-BA treatment can improve the activity of lotus SOD,and the concentration of 150mg/L 6-BA is the best.150mg/L 6-BA and120mg/L GA3 treatment maximizes POD activity and enhances POD activity,which effectively extending the viewing period of lotus flowers.120mg/L GA3 treatment inhibits the increase of MDA activity,which can prolong the aging of lotus petals and prolong the ornamental period of lotus.CAT activity showed the highest activity during the blooming period from the budding stage to the flowering-wilting stage gradually decreased.The gibberellin is the best in three kinds treatment of exogenous hormone.(3)Study and analysis of related genes of lotus petals senescence in different treatments.GA3 treatment significantly promoted the expression of EXP genes in the initial flowering period and full-flowering stage,which the expression quantity of EXP gene was higher than that of other treatments.The expression of EXP gene was inhibited by 150mg/L GA3 treatment.150mg/L 6-BA and 150mg/L GA3 was used to promote the expression of Cu Zn SOD gene of lotus petals,and all of them peaked during the fulling-flowering period.The treatment of 120mg/L GA3 and 150mg/ L 6-BA can inhibit the expression of CKX gene during the growth and development of lotus,which is more conducive to the growth of lotus,and has a significant extension of flowering stage.
