For the rapid promotion of 全haoqiu No.3'Platanus acerifolia,In this paper,a rapid propagation system was established based on the three aspects of the hardwood and softwood cutting technology and the Plant tissue culture.The results are as follows:(1)The experiment on the tissue culture system of the全haoqiu No.3' Platanus acerifolia: when the stem is explant,the buds in the middle of the branches have higher survival rate and are more suitable to sterilize and induce germination.The best sterilization scheme for the stem explants are 75%Alcohol 30s+2%Naclo(10min)+0.1%Hgcl2(8min)and 75%Alcohol 30s+2%Naclo(10min)+0.1%Hgcl2(6+6min);The optimal sterilization scheme for leaves is 75%Alcohol 20 s + 0.1%Hgcl2(4+6)min.The optimum culture medium for the germination of axillary buds is MS+IBA 0.3mg/L+6-BA 1.5mg/L;The optimum culture medium for leaf blade induced adventitious buds is MS + 6-BA 2mg/L+IBA 0.3mg/L;Dark culture can shorten the differentiation time of adventitious buds and reduce the browning of callus,The best medium for bud proliferation and callus induced adventitious buds is MS+6-BA 1.0mg/L+ IBA 0.5mg/L +NAA0.1mg/L;The optimal medium for rooting is 1/2MS+IBA0.1mg/L.(2)The survival rate and the number of leaves and rooting of hardwood cutting in the six years maternal plant are significantly higher than that in the 16 years,the survival rate and the leaves number in the six years maternal plant that be saved in short branch over winter was highest in all processing.The long and short branches with NAA over winter had the highest survival rate when NAA was 100mg/L,and the survival rate of short branches was greater than that of long branches.200mg/LNAA can promotes the germination length of short branches,and the length of germination branch of short branches is larger than that of long branches.The average number of rooting in the short branch with 100 mg/L NAA over the winter was the highest in all processing,and the average number of rooting in the short branches was greater than that of the long branches.In the middle of June,the survival rate of softwood cuttings was highest,and the survival rate,rooting number and average rooting length of cuttings from the six years maternal plant were more than 16 years.The survival rate and rooting number of cuttings with one leaf and two buds were larger than that of cuttings with one leaf and three buds,and there was no obvious correlation between the root length and the cutting methods.The survival rate of cuttings with one leaf and two buds from the six years maternal plant was the highest in all treatments.The survival rate and rooting number of cuttings in lighting condition were greater than that of cuttings in the shade condition.The rooting number of cuttings with one leaf and two buds is greater than that of cuttings with one leaf and three buds,the average rooting length of cuttings in the shade condition is higher than that of cuttings in the lighting condition,and the average rooting length of cuttings with one leaf and two buds were the largest.The relationship between the average root length and NAA concentration was not obvious.Under full lighting,The survival rate of cuttings with one leaf and two buds was the highest in 300mg/LNAA,200mg/L NAA was favorable for the survival rate of cuttings with one leaf and three buds;Under the shade,200mg/L NAA was favorable for the survival rate of cuttings with one leaf and two buds,and 300mg/L NAA was favorable for the survival rate of cuttings with one leaf and three buds.The rooting length and number of cuttings in vermiculite and peat soil(1:1)mixed matrix are greater than that of in the sand and vermiculite(2:1)mixed matrix,but the rooting roughness is less than the mixture of sand and vermiculite(2:1).
