In order to further enrich and improve the Idesia Polycarpa vegetative propagation technology system, the studies used robust and excellent mother tree of Idesia Polycarpa from experimental field as test materials, and designed the experiments of hardwood cutting, root burying, softwood cutting and tissue culture, researched the effects of cuttings length(6 cm,8 cm,10 cm), concentrations of ABT(0 mg/kg, 2000 mg/kg) and existence of shading on hardwood cuttings, researched the effects of root length(5 cm,10 cm), root diameter(0.1-0.5 cm,0.5-1.0 cm), concentrations of ABT(0 mg/kg, 2000 mg/kg) and root buried ways(horizontal buried,vertical buried) on root burying, researched the effects of cutting period(6-15,6-30,7-10,7-25,8-10,8-26), cuttings positions(the tip of branch, the following tip of branch), amounts of leaves reserved(whole leaf, half leaf) and concentrations of ABT(0 mg/kg, 500 mg/kg, 1500 mg/kg, 2000 mg/kg) on softwood cutting, and screened the mediums of inducing-callus, inducing-buds and inducing-rooting. The results showed that:(1) The best treatment of hardwood cutting is J6 which owns the highest survival rate, it was that made Idesia Polycarpa branches to 6 cm length cuttings, then speedly dipped the cuttings for 30 s in the number 1 of ABT with a concentration of 2000 mg/kg, and the survival rate was up to 79.33%.(2) The best trearment of root burying is CS10 which owns the highest survival rate, namely chosen the root diameter of 0.5-1.0 cm and length of 10 cm, vertical buried without ABT, the survival rate was up to 98.00%.(3) For the thick roots in the length of 10 cm,vertical buried effect is better. And used the thick roots in the length of 5 cm to bury is feasible by ABT process and horizontal buried way, the survival rate was up to 90.33%. For the fine roots, horizontal buried effect is better, cut the fine root to the length of 10 cm, then buried it horizontally after ABT treatment, the survival rate was up to 90.67%.(4) The best treatment of softwood cuttings of Idesia Polycarpa is the 5th treatment in the period of July 25 th, namely chosen the following tip of branch as cuttings without ABT treatment and reserved one piece of whole leaf, the callus rate could reach 100 % and rooting rate could reach 41.83 %.(5) 75% alcohol 30 s + 0.1% Hg Cl2 6 min was the best disinfection for leaves, the contamination rate was 25% and the browning rate was 18%; 75% alcohol 30 s + 0.1% Hg Cl2 8 min was the best disinfection for buds, the contamination rate was 13% and browning rate was 12%; 75% alcohol 30 s + 0.1% Hg Cl2 10 min was the best disinfection for stem, the contamination rate was 10% and browning rate was 17%. With the increase of immersion time under 0.1% Hg Cl2, explants pollution rate decreased, but it was easy to damage explant, causing browning.(6) The best medium for callus induction of Idesia Polycarpa was 1/2MS + TDZ 0.02 mg/L + 6-BA 1.0 mg/L + NAA 0.2 mg/L, inducing 98%.(7) The best medium for rooting induction of Idesia Polycarpa was 1/2MS + IBA 2.0 mg/L + NAA 0.05 mg/L, the induction rate was up to 100%, the average amount of new root was 12 and the average length of new root was up to 4 cm.
