Hu sheep is a unique lamb skin-type sheep variety of China.With high breeding rate and tender meat,Hu sheep matures early,therefore it can be breeding all year round.However,comparing with the Dorper sheep and other sheep specialization varieties,the meat performance and dressed percentage of Hu sheep is relatively low.This paper is for muscle characteristics of male born and six month old Hu sheep and F1 Dorper x Hu sheep,and the expression of MYL3 and ACTG2 gene variable splicing in different muscle tissues to study.To study the molecular mechanism of skeletal muscle growth and development of Hu sheep.To explore the relationship between MYL3 and ACTG2 gene with the muscle fiber growth and fat accumulation,which can provide new thinking for the breeding of new strains of meat sheep and lay a theoretical basis for the breading of new variety of meat-type Hu sheep.The test slaughtered 12 male born and six month old Hu sheep and F1 Dorper x Hu sheep(newly born,6 month old each half)?and each selected BFM,LDM,MGM,GM,VLM,SOL,SSC 7 muscle tissue:(1)Using paraffin sections,HE staining method were measured the diameter and length of muscle fibers in different muscle tissues.The fat distribution of muscle samples was shown by oil red O staining.(2)Analyzed the expression of MYL3,ACTG2 and it 5'UTR variable splicing in 7 kinds of muscle tissue of male Hu sheep,Dorper x Hu sheep.(3)Construction the Double Luciferase Reporter gene of ACTG2 5' UTR variable splicing,and transfection Porcine Kidney cells(PK-15)and Chicken Fibroblasts cells(DF-1)to detection the luciferase activity and the expression of ACTG2 5' UTR variable splicing.The results are as follows:(1)Except male Hu sheep muscle diameter and length there were no significant differences between the newly born sheep and the 6 month old sheep.The diameter and length of BFM,LDM,MGM,GM,VLM and SSC of male newly born Hu sheep were significantly or significantly less than that 6 month old sheep(P<0.05 or P<0.01).The muscle fibers diameter of the LDM in newborn and 6 month old male Hu sheep was significantly less than the same month Dorper xHu sheep(P<0.05).The muscle fibers length of the BFM(P<0.05)?LDM(P<0.01)in male 6 month old Hu sheep was significantly or high significantly less than the same month Dorper xHu sheep.The IMF content in BFM of male mewly bom Hu sheep and Dorper xHu sheep was significantly less than their 6 month old sheep(P<0.05),and the IMF content in SOL was significantly less than the same variety 6 month old sheep(P<0.01).The IMF content in VLM.SSC of male mewly born Hu sheep was significantly less than the same variety 6 month old sheep(P<0.05).(2)Construction the Double Luciferase Reporter gene pLUC-65.pLUC-83?pLCUC-116 of ACTG25'UTR variable splicing(65bp.83bp.116bp),and the relusts of transfected Porcine Kidney cells(PK-15)and Chicken Fibroblasts cells(DF-1)were shown:The activity of pLUC-83 promoter is the strongest,can significantly increased the expression of vector of ACTG2 5r UTR 83bp sequence.(3)The expression of MYL3 in SOL of newly born male Hu sheep significantly higher than that in LDM?SSC(P<0.01),and significantly higher than that in MGM(P<0.05);The expression of MYL3 in BFM significantly higher than that in LDM,SSC(P<0.05);The expression of ACTG2 5' UTR 83bp splicing in SOL significantly higher than that in LDM(P<0.05),and it proportion of total expression of ACTG2 in SSC was significantly higher than that in BFM,GM,VLM and SOL(P<0.05).The expression of MYL3 in BFM of 6 month old male Hu sheep significantly higher than that in the other six tissues(P<0.01),and it expression in LDM of significantly higher than that in SSC(P<0.05);The expression of ACTG2 in BFM?MGM significantly higher than that in the other five tissues(P<0.05);The expression of ACTG2 5‘UTR 83bp splicing in VLM significantly higher than that in LDM?MGM?SSC(P<0.05);The expression of ACTG2 5' UTR 83bp splicing inLDM of newly born Hu sheep significantly higher than that in BFM?MGM(P<0.01),and significantly higher than that in GM?VLM?SOL(P<0.05);The expression of MYL3 in BFM of 6 month Dorper ×Hu shep significantly higher than that in LDM.VLM?SSC(P<0.05);The expression of ACTG2 in MGM significantly higher than that in BFM(P<0.01),and significantly higher than that in GM?VLM?SSC(P<0.05);The expression of ACTG2 5' UTR 83bp splicing in SOL significantly higher than that in the other six tissues(P<0.01),and it expression in LDM significantly higher than that in MGM?GM?VLM(P<0.05).MYL3?ACTG2 gene and ACTG25 "UTR 83bp splicing have a certain impact on the muscle growing development and meat quality of Hu sheep.
