| Banana(Musa spp.)is one of the most important fruit and food crops.Fusarium wilt caused by Fusarium oxysporum f.sp.cubense(Foc)is the most destructive diseases substantially reducing the production of banana in the world.There is still no effective way to prevent it.Studying the disease resistance mechanism of bananas and cultivating new varieties of disease resistance are very important tasks.In this experiment,the RNA-seq technique was used to analyze the differences expression genes between the resistant variety ’Dongjiao 1’(Musa spp.AAA)and its susceptible strain Ke2(Musa spp.AAA)under Foc 4 stress tolerance.The analysis of the disease resistance mechanism of bananas by GO(Gene ontology)enrichment and KEGG(Kyoto encyclopedia of genes and genomes)pathway analysis provides a theoretical reference for banana disease resistance breeding.The tests of DUS of three new clones and disease resistance was investigated on two new clones.so as to lay the foundation for cultivating new varieties of banana with good traits.The main results obtained are showed as follows:1.RNA-seq analysis(1)1 There were 5118 differentially expressed genes(DEGs)under Foc4 stress,and 3466 up-regulated expression.Of these DEGs,70 DEGs were only expressed in pathogenic bacteria infected ’Dongjiao1’(DJI-DJCK);31 DEGs were significantly more highly expressed in ’Dongjiao1’ infected by pathogenic bacteria than in Ke2(DJI-Ke2I);and there are 6 DEGs were not only induced in DJI-DJCK comparison group,but also up-regulated in DJI-Ke2 I comparison group.Suggesting it involvement of the disease resistance.,suggesting their involvement of the disease resistance.(2)The differentially expressed genes obtained under Foc4 stress can be annotated to 2073 GO items.There are 740 GO items,1669 items and 294 items for the molecular function,biological process and cell components.In the molecular function GO items,14 GO items that are only enriched in the comparison of DJI-DJCK.There are 8 GO items that are only enriched in the comparison of DJI-Ke2 I.And there are 4 GO items simultaneously enriched in the comparison of DJI-DJCK and DJI-Ke2 I.In the biological process,14 GO items that are only enriched in the comparison of DJI-DJCK. There are 5 GO items that are only enriched in the comparison of DJI-Ke2 I.1 GO item was not only enriched in the comparison of DJI-Ke2 I but also in the comparison of DJI-DJCK,suggesting their involvement of the disease resistance.(3)The differentially expressed genes obtained under Foc4 stress can be annotated to 107 KEGG pathway.1 KEGG pathway was only enriched in the comparison of DJI-DJCK. Suggesting that the up-regulated genes in the KEGG pathway may be related to the disease resistance of bananas.2.The main results of DUS test(1)The leaf spacing of the three new clones was greater than that of the control cultivar.The leaf spacing of HNAJ0109 was significantly greater than that of the ‘Beidaijiao’ (Musa spp.AAA),and its leaves were also significantly wider than ‘Beidaijiao’.The stem length of the new clones was longer than that of the ‘Beidaijiao’,and the length of the stem of the HNAJ0212 was the longest(44.35 cm),which was significantly longer than that of the ‘Beidaijiao’.The ear length of each variety(clone)was between 69.20 cm and 77.43 cm.The ear length of the new clones,HNAJ0109,was the longest, which was significantly longer than that of ‘Beidaijiao’.(2)‘Beidaijiao’ and HNAJ0212’s neutral flowers have a strong presence,HNAJ0109 does not exist,and HNAJ0301 is among them.(3)The edible rates of the three new clones were all significantly greater than those of the‘Beidaijiao’.3.Identification of disease resistanceAfter 28 days of inoculation,the disease index of HNAJ0107 and HNAJ0208 was between the susceptible variety ’Baxijiao’(Musa spp.AAA)and the disease resistant variety’Yueyoukang 1’(Musa spp.AAA).It is speculated that the two new strains may have certain disease resistance. |
