Cymbidium Jade Hare is a grex of Cymbidium sinense and Cymbidium lancifolium,several hybrid Cymbidium sinense cultivars including Cymbidium Jade Hare‘Yuxiang'and Cymbidium Jade Hare‘Yuyuan'have been created from the grex.These cultivars with the advantages of both parents has high ornamental value and broad market prospect.At present,there are some reports about tissue culture and rapid propagation of Cymbidium sinensis,but there are few reports about the micropropagation technology of hybrid Cymbidium sinensis seedling.In this paper,Cymbidium Jade Hare‘Yuxiang'and Cymbidium Jade Hare‘Yuyuan'were employed to investigate micropropagation characteristics of hybrid Cymbidium sinensis and the industrial production technology of Cymbidium Jade Hare‘Yuxiang'seedling.The main research results are as follows:1.Micropropagation characteristics of Cymbidium Jade HareCymbidium Jade Hare‘Yuxiang'and Cymbidium Jade Hare‘Yuyuan'were easier for micropropagation compared with Cym.‘Qijianbaimo'.The rhizome induction rate of Cymbidium Jade Hare‘Yuxiang'and Cymbidium Jade Hare‘Yuyuan'was 38.6%and 35.3%,respectively,which was higher than that of Cym.‘Qijianbaimo';the proliferation coefficiency was 1.91 and 2.02,respectively,which was significantly higher than that of Cym.‘Qijianbaimo',while there was no significant difference in browning rate;The root and shoot regeneration rates were 20.83%,64.58%and 14.17%,37.92%,respectively,which was significantly higher than that of Cym.‘Qijianbaimo'?0.42%,0.83%?.The growth of seedlings was faster,and the survival rate and commercial rate of seedlings were higher compared with Cym.‘Qijianbaimo'.2.Factors influencing on efficiency of micropropagation in Cymbidium Jade Hare‘Yuxiang'Basic medium,plant growth regulator,activated carbon and dark condition had a significant effect on rhizome proliferation of Cymbidium Jade Hare‘Yuxiang'.Liquid suspension culture significantly decreased the multiplication coefficient,but supplement of2.0mg·L-1 Cu2+significantly increased the multiplication coefficient.The proliferation coefficient was decreased by cutting treatment,and there was a significant effect on proliferation of the rhizome by supplement of graphene oxide.Adding 150.0 mg·L-1 coconut water increased the multiplication coefficient.The suitable medium for rhizome proliferation was MS+6-BA 1.0 mg·L-1+NAA 0.5 mg·L-1+AC 0.5 g·L-1+Cu2+2.0 mg·L-1+coconut water150.0 ml·L-1+sucrose 30.0 g·L-1+carrageenan 7.0 g·L-1?pH=5.8±0.2?,on which,the proliferation coefficient was 2.65.Basic medium,plant growth regulator and activated carbon had significant influence on rhizome differentiation of Cymbidium Jade Hare‘Yuxiang'.The root differentiation rate and mean number of root regenerated were improved,however,the bud differentiation rate,mean number of bud regenerated decreased by adding 80 g·L-1 banana;the bud differentiation rate,mean number of bud regenerated were improved,however,the root differentiation rate and mean number of root regenerated decreased by adding Brassinolide and methyl jasmonate,but adding 6 mg·L-1 Cu2+significantly increased the bud differentiation rate,mean number of bud regenerated,root differentiation rate and mean number of root regenerated.The suitable medium for rhizome differentiation was MS+6-BA 1.5 mg·L-1+NAA 0.5 mg·L-1+AC 0.05 g·L-1+Cu2+6 mg·L-1+sucrose 30 g·L-1 and cara powder 7 g·L-1?pH=5.8±0.2?,on which,the bud and root differentiation rates were 73.68%and 35.62%,respectively.Basic medium and plant growth regulator had significant influence on seedling growth,0.5 g·L-1 active carbon promoted seedling growrh of Cymbidium Jade Hare‘Yuxiang'.The suitable medium for seedling growth was MS+6-BA 0.1 mg·L-1+NAA 0.5 mg·L-1+AC 0.5g·L-1+sucrose 20 g·L-1+carrageenan 7 g·L-1?pH=5.8±0.2?,the height of 4-cm-height seedling reached 7.25 cm aftercultured on the medium for 60 d.The survival rate of 8-cm-height seedling was 99.0%after transplanting 90 d.Through the above study,micropropagation characteristics of hybrid Cymbidium Jade Hare was clarified,factors influencing on micropropagation of Cymbidium Jade Hare‘Yuxiang'were elucidated,and the technology system for rapid propagation by tissue culture was established,which laid a technical foundation for factory production of Cymbidium Jade Hare‘Yuxiang'seedling.
