Toxoplasma gondii(T.gondii)can infect almost all the nuclear cells of warm-blooded animals.In the world,the number of Toxoplasma gondii infections is more than 30% of the total population.Infections via mouth,blood,placenta are all important ways of T.gondii.The rate of pregnancy abnormalities of women infected with T.gondii during pregnancy is three times higher than normal pregnant women's.The main adverse effects include infertility,abortion,fetal reabsorption,stillbirth and the ability of the carrier to produce cysts.T.gondii can influence the pregnancy outcome through a variety of ways such as directly damaging uterus,intrauterine environment,hypothalamus and hormone balance.The RH strain is an international standard type of T.gondii type I stain,to which mouse was highly sensitive.After 72 h of T.gondii infection,this parasite can be detected in almost all kinds of cytoplasm in the uterus.The transcriptome sequencing can help researchers obtain the gene expression status and the regularity of transcriptional regulation of organisms.So that the development of the body and the process or the potential pathway of disease occurrence can be fully understood.In this study,we could find the link between differentiating pathways and complications.We could also find genes that cause early pregnancy failure by RNA-Seq binding bioinformatics analysis.This study aimed to find the key genes and pathways of the adverse pregnancy outcome of Toxoplasma gondii infection,and how tachyzoite interferes the genes involved in gestation.As treatment groups,we used tachyzoite to infect female mice on different days of pregnancy,which included third days before pregnancy,third days after pregnancy,and third days after implantation.Meanwhile,we used the normal saline to set up control groups.Transcriptome sequencing was performed on the uterine samples of the mice after 3 days of infection.The three groups were detected 4561,2345 and 2997 significant differentially expressed genes(DEGs),respectively.And we used GO annotation and KEGG pathway analysis to analyze differentially expressed genes.The DEGs were related to anatomical structure development,transport,cell differentiation,embryo development,hormone biosynthetic process,and immune response.And,the DEGs were mainly located in three pathways,including Autophagy,Cancer,and Cytokine-cytokine receptor interaction.Among them,the Cancer Pathways were the most significant difference pathways in the three experimental group.The change of Wnt signaling Pathway was the most significant in it.In previous studies,the classical Wnt /?-catenin pathway has a close relationship with embryo and embryo development.We found many factors that T.gondii affect hosts' pregnancy outcomes,including the gestational hypertension,the destruction of the intrauterine environment,the obstacles of the placental formation and function,and the effect of the embryo development.Through further analysis,we screened out DEGs that are closely related to the pregnancy and T.gondii,such as Hsd11b2,Ifitm1,CD74,CXCL14,Ptges,etc.When the T.gondii infects hosts,the changes of these mRNA probably cause the pregnancy failure.Our study may help identify the key genes involved in the adverse pregnancy after T.gondii infection.In addition,the study will help to better understand the molecular mechanism of the acute infection of the Toxoplasma gondii,and to further develop a new treatment for toxoplasmosis in pregnancy.
