Oxidative stress refers to the imbalance of oxidation and antioxidation in animals.It is a negative effect of the long-term overproduction of free radicals in animals.It is considered to be one of the important factors leading to animal aging and disease.During animal breeding,oxidative stress can cause animal diseases such as heart disease,kidney disease,ascites,perinatal diseases,retention of placenta,mastitis,and digestive tract inflammation,which in turn affect animal production and threaten the healthy development of the aquaculture industry.Oxidative stress also induces the expression of cyclooxygenase?COX?and lipoxygenase?LOX?,promotes the secretion of inflammatory mediators,induces inflammatory reactions in animals and reduces animal performance.Apigenin-7-O-glucoside?AG?is the main active flavonoid in the water extract of Chamomile.AG is considered to be one of its most abundant components,and it has antioxidant and anti-oxidant properties.The inflammatory activity,its antioxidative and anti-inflammatory activity has not yet been fully elucidated.This article has carried on the preliminary study of AG anti-oxidation,anti-inflammatory activity,in order to provide the basis for AG in the veterinary clinical application.In this thesis,1,1-diphenyl-2-picrylhy-drazyl?DPPH?and ferricion reducing antioxidant power were used.?FRAP?,preliminary evaluation of the antioxidant capacity of AG in the chemical system,found that AG capture DPPH free radicals and Fe3+reducing ability is weak;through 2,2'-azobis?2-amidinopropane?hydrochloride?AAPH?induces oxid ative damage of red blood cells and plasmid pBR322,and Cu2+/H2O2 induces oxidative damage of bovine serum albumin?BSA?.Antioxidant capacity in the biological system,the results found that AG in the above system can be a good reduction of red blood cells in sheep,plasmid pBR322and BSA free radical oxidation damage,showing good antioxidant activity.In this thesis,RAW264.7 cells were stimulated with lipopolysaccharide?LPS?to construct an inflammatory model,and the anti-inflammatory activity of AG was evaluated.It was found that AG can significantly reduce the production of nitric oxide?NO?in RAW264.7 cells and reduce the tumor necrosis factor-??TNF-??and interleukin-6?IL-6?.secretion.Reverse transcriptase-polymerase chain reaction?RT-PCR?studies revealed that AG can also inhibit RAW264.7 cell mRNA expression of intracellular of inducible Nitric oxide synthase?iNOS?,TNF-?,and IL-6 by LPS-induced through reverse transcriptase-polymerase chain reaction.It was shown that AG can well alleviate the inflammatory response produced by LPS-stimulated RAW264.7 cells and has good anti-inflammatory activity in this system.
