| Objective: Glycyrrhiza polysaccharide(GP)is polysaccharides extracted from the natural plant.It has been proved to have immune promoted functions,anti-tumor,anti-cancer,anti-oxidation,antibacterial,anti-inflammatory and antiviral,which can be developed as functional feed additives with no residues and no toxicity.The purpose of the experiment is to study the immunomodulatory,the antioxidant and the anti-inflammatory activity of Selenizing Glycyrrhiza uralensis Fisch polysaccharide(SeGUP)on the immune function,anti-inflammation activity and antioxidant capacity of Jinghong No.1 chicks,provide theoretical basis for the application of SeGUP in layer breeding.Methods: 300 one-day-old Jinghong No.1 chicks were randomly divided into 5 groups: control group,LPS group,LPS+Astragalus polysaccharides(APS)group,LPS+Glycyrrhiza uralensis Fisch polysaccharide(GUP)group and LPS+SeGUP group.Intraperitoneal injection of LPS was used to establish a chick immune stress model.After the immune stress model was established by intraperitoneal injection of LPS on the 8th,10 th and 12 th day of the test,the test group was injected with APS,GUP and SeGUP for 7 days,and samples were taken on the 7,14,21,28,and 35 days after the administration.,phagocytic index were measured by carbon particle clearance method,the serum levels of IgA,IgG,IgM,C3,C4,s CD4,s CD8,TNF-α,IL-1β,IL-2,IL-6,NO,iNOS and intestinal mucous s IgA were determined by enzyme immunoassay,the contents or activities of T-AOC,SOD,GSH-Px and MDA in serum,liver and kidney tissues were determined by biochemical method.Results:(1)SeGUP significantly inhibited the increase of macrophage phagocytic index in 18,25,32,39 and 46-day-old chicks stimulated by LPS(P<0.05).SeGUP significantly decreased the content of IgA in serum of 25-day-old chicks stimulated by LPS(P<0.05)and the contents of IgG and IgM in serum of 18,25,32-and 39-day-old chicks(P<0.05),increased the content of s CD4 in serum of 18,25-and 32-day-old chicks stimulated by LPS(P<0.05),and decreased the content of s IgA in intestinal mucous of 18,25,32,39-and 46-day-old chicks stimulated by LPS(P<0.05).It was suggested that SeGUP alleviates the immune challenge response induced by LPS stimulation in the immune system and plays an immunomodulatory role.(2)SeGUP significantly increased the serum T-AOC content of 18 and 25-day-old chicks stimulated by LPS(P<0.05),the serum SOD activity of 18,25,32,39-and 46-day-old chicks(P<0.05),and the serum GSH-Px activity of 18,25,32-and 39-day-old chicks(P<0.05),while significantly decreased the serum MDA content of 18,25,32,39-and 46-day-old chicks(P<0.05).It significantly increased the content of T-AOC in the liver of 39-day-old chicks(P<0.05),the activity of SOD in the liver of 32-and 46-day-old chicks(P<0.05),and the activity of GSH-Px in the liver of 18,25-and 46-day-old chicks(P<0.05).It significantly decreased the content of MDA in the liver of 18,25,32,39-and 46-day-old chicks(P<0.05).It significantly increased the content of T-AOC in the kidney of 18,32,39 and 46-day-old chicks(P<0.05),increased the activity of SOD in the kidney of 18,25,32-and 46-day-old chicks(P<0.05),significantly increased the activity of GSH-Px in the kidney of 18,25,32-and 46-day-old chicks(P<0.05),and decreased the content of MDA in the kidney of 18,25,32,39-and 46-day-old chicks(P<0.05).It is suggested that SeGUP can alleviate the stress caused by LPS by enhancing the activity of antioxidant enzymes in chicks.(3)SeGUP significantly alleviated the increase in serum levels of IL-1β,IL-2,IL-6,TNF-α,NO and iNOS contents in 18,25,32,39 and 46-day-old chicks caused by LPS stimulation(P<0.05).It was suggested that SeGUP could alleviate the inflammatory response caused by LPS stimulation in chicks,and play an anti-inflammatory effect.Conclusion: SeGUP could alleviate the immune stress caused by LPS and improve the antioxidant capacity of chicks,thereby exerting immunomodulatory,anti-inflammatory and antioxidant effects. |
