This article is divided into four parts to study the effects of sodium butyrate and Artemisia seed polysaccharide on growth performance,intestinal development and glucagon-like peptide-2 in weaning lambs.In the experiment,48 Ordos wool sheep with a similar age and weight were selected and divided into four groups(n=12).Control group(CG)?Sodium butyrate group(SB)?Artemisia seed polysaccharide group(ASP)?Sodium butyrate + Artemisia seed polysaccharide group(SB+ASP).The trial period is 36 days.Part I Effect of Sodium Butyrate and Artemisia seed Polysaccharide Added to Diet on the Growth and Development of Weaning Lambs.This section first tests the fitness of the diet.The weight of the lamb is weighed exactly before the morning feeding every 12 days during the test period.Calculate weight change and average daily gain,At the end of the trial,3 lambs were randomly selected from each group for slaughter,Calculate live weight before slaughter,carcass weight,organ weight,and gastrointestinal weight,and measure the length of the small intestine.Diet fitness test analysis found Artemisia polysaccharides group has the highest positive combination effect,Sodium butyrate group has the lowest positive combination effect.Sodium butyrate significantly increased average daily gain(P< 0.05),and Artemisia seed polysaccharide also significantly increased average daily gain(P< 0.05),Sodium butyrate plus Artemisia seed polysaccharide also increased the average daily gain but did not differ significantly from the sodium butyrate group and the Artemisia polysaccharide group.This group did not show a combination effect.Sodium butyrate and Artemisia seed polysaccharides can significantly increase the weight of spleen Sodium butyrate can significantly increase the weight of the rumen and abomasum(P< 0.05).And sodium butyrate can significantly increase the weight of the small intestine(P<0.05).Artemisia seed polysaccharides group can also significantly increase the weight of small intestine(P< 0.05).Part II Effect of Sodium Butyrate and Artemisia seed Polysaccharides Added to Diets on Intestinal Villus Morphology of Weaned Lambs.This part of the experimental animals and feeding management with the first part After the end of the trial,3 lambs were randomly selected from each group for slaughter.Duodenum,jejunum and ileum were used for hematoxylin-eosin(HE)staining.Measure the ratio of intestinal villus height,crypt depth,and intestinal villi height to crypt depth(V/C).The results showed that the sodium butyrate group,the Artemisia seed Polysaccharides group,the sodium butyrate + Artemisia seed Polysaccharides group all significantly increased the intestinal villus height(P< 0.05),and increased the ratio of intestinal villi height to crypt depth(V/C).All can improve intestinal villus morphology and promote nutrient absorptionPart III Effect of Sodium Butyrate and Artemisia seed Polysaccharides Added to Diet on Blood Biochemical Indexes of Weaning Lambs.This part of the experimental animals and feeding management with the first part,During the trial,blood was collected from the jugular vein on the first,fourth,eighth,12 th,24th and 36 th days of the test.After serum collection,relevant indicators were measured.The results of the trial showed that the levels of serum IGF-1 in the sodium butyrate and Artemisia seed polysaccharides group were significantly higher than those in the control group.Artemisia seed polysaccharides can significantly increase serum levels of nitric oxide(NO)and nitric oxide synthase(i NOS)(P< 0.05),Moreover,sodium butyrate and Artemisia seed polysaccharides can significantly reduce serum C-reactive protein(CRP)and serum amyloid A(SAA)levels(P< 0.05).In addition,the levels of glucagon-like peptide-2(GLP-2)in the sodium butyrate and Artemisia seed polysaccharides groups during the pre-test period were significantly higher than those in the control group(P< 0.05).Part IV Effect of Dietary Sodium Butyrate and Artemisia seed Polysaccharides on Expression of Related Proteins in Weaning Lamb Small Intestine.This part of the experimental animals and feeding management with the first part,Three lambs were randomly selected from each group to be slaughtered at the end of the trial.The duodenum,jejunum and ileum were taken.Western blot analysis of the expression of relevant proteins in the intestine The results showed that sodium butyrate and Artemisia seed polysaccharides increased the expression of Occludin,Caludin and ZO-1 proteins,All can improve intestinal barrier function.Sodium Butyrate and Artemisia seed Polysaccharides Can Promote Secretion of GLP-2 in Intestine.Sodium butyrate and Artemisia seed Polysaccharides in the jejunum and ileum are likely to increase SIRT-1 expression.In the jejunum and ileum,sodium butyrate is likely to be involved in two transcription factors,NF-?B and P53,and Artemisia seed polysaccharide is likely to involve two transcription factors,Fox O1 and P53.
