Cercis glabra,a genus of leguminous bauhinia,also known as bamboo baskets,is an economical,ornamental,and medicinal species with high value.But seed germination characteristics of seed coat is hard and long period,cutting propagation of cuttings are hard to survive,the west of matrix and soil affect the growth of seedlings,severely restricts the application of Cercis glabra.In addition,there are few reports on the related breeding of bauhinia in Cercis glabra,especially the research on cutting propagation technology,rooting mechanism and matrix screening is blank.Therefore,it is of great significance to explore the breeding technology for the industrial production and good germplasm protection of bauhinia seedlings in Cercis glabra.This test by studying the seeding method,cutting method and physiological characteristics,the west matrix filter,etc.,in order to select the optimal breeding methods,to provide theoretical reference for the breeding of Cercis glabra application.The research results are as follows:(1)Cercis glabra is a hard seed.After germination for 10 min with concentrated sulfuric acid and 24 h with gibberellin,the germination rate is best at 25 ?,and the germination rate is 83.21 %.(2)In the mid-August,when cuttings were selected,they were planted with a 5-cm long,semi-lignified cutting with two nodes and treated with ABT 1000 mg/L or NAA 1000mg/L hormones.The rooting rate was the best in the matrix with =3:1,and the rooting rate was up to 26.59 %.(3)The rooting type of the Cercis glabra is inducible and does not contain root primordia.The water content,soluble sugar,soluble starch,C/N,soluble protein,total phenol,CAT decreased,total nitrogen,POD,PPO,SOD,MDA,PRO increased at the initial stage of cutting;water content,soluble starch,total nitrogen,adventitious root formation period,Total phenol and CAT decreased,soluble sugar,C/N,soluble protein,POD,PPO,SOD,MDA,PRO increased;soluble sugar,C/N,soluble protein,POD,PPO,SOD,MDA,PRO decreased after adventitious root formation,Water content,soluble starch,total nitrogen,total phenol,CAT rose.Hormone-treated cuttings showed significantly shorter change cycles than untreated cuttings.Hormone treatment improved the utilization of water content,nutrients,and total phenols,increased the activity ofoxidase and improved its resistance.Viability eventually promotes rooting.(4)In the matrix screening of seedlings,the minimum physical weight of T1 was 0.14g·cm-3,the maximum total porosity was 82.52 %,the maximum pore volume was 31.68 %,and the maximum pore size was 50.84 %.The size of the pores was excellent at 0.62.According to the chemical properties of the substrate,the highest nitrogen of T1 was170.52 mg·kg-1,the highest available phosphorus was 85.05 mg·kg-1,and the highest available potassium was 289.69 mg·kg-1.The lowest pH value was 6.13,and the highest EC value was 807.10 us·cm-1.The effect of substrate on growth index of seedlings was obtained.The length of T1 leaf was 6.7 cm,the width of leaf was 4.6 cm,and the maximum was 20.9 cm.The maximum crown width was 18.1 cm and the maximum number of leaves was 11.The chlorophyll content of T1 was 21.12 mg·g-1,the highest water content was 80.47 %,and the minimum conductivity was 15.12 %,the highest soluble sugar content was 3.85 %,the highest soluble protein content was 321.37 ?g·g-1,the highest SOD activity was 115.87 ?g-1·Fw,the highest POD activity was 61.67?g-1·min-1·Fw,and the lowest was MDA It is 5.246 ?mol·g-1 and PRO is the highest at25.84 ?g·g-1,so T1 is most suitable for Cercis glabra.
