The Haliotis discus hannai is attached to Gastropoda,widely distributed along the coast of northern of China,such as Liaoning and Shandong province.It has become one of the most important economic species in China as its delicious taste and high nutritional value.However,with the deterioration of breeding environment and the decrease of genetic diversity in recent years,the healthy breeding and sustainable production of abalone are facing great challenges,so it is imperative to improve the variety of abalone.The abalone was used as the research object in the study,and there are three parts in the study: Firstly,the genetic distance of 31 candidate parents was estimated based on microsatellite markers,and selected the parents to construct the green eggs family based on the result.Next,compared the growth and survival among green family for finding the dominant family.Then observed the species and the proportion of egg color of each family.Finally,the types and contents of pigments in the gonads of male and female abalone were studied based on the high performance liquid chromatography(HPLC).In order to clarify the genetic mechanism of egg color of abalone and to provide theoretical support for breeding of abalone abalone,the correlation between egg color and survival status of abalone and inheritance of egg color of Haliotis discus discus were discussed in combination with the growth status of different families in order to find out the genetic mechanism of egg color of abalone.It provides theoretical support for the breeding work of H discus hannai.The main findings are as follows:1.The selection of candidate parents for the Green Egg family of the abaloneThe genetic distance between 11 female and 20 male of abalone was estimated based on the microsatellite marker technique.According to the results,we selected two females and two males to construct the green egg family.2.The establishment of green egg family and the comparison of growth and survival According to the result,we selected the parents of the green egg family.We also selected three non-green-egg females and three male of abalone as the parents of the non-green egg family.The selected parents were used to establish four full-sibs green egg family and a half-sibs non-green egg family.The results show that the body length,body width and body weight of the green eggs were increased but the family 1 showed the biggest growth advantage in different growth periods,and compared with otherfamilies,the family 2 showed the excellent characters of fast growth rate.In addition,we have verified the conclusion that its no absoluteiy connected between the genetic distance of parent and the growth traits of the offspring.3 Analysis on the genetic regularity of the egg colorThe F1 gonad was treated by oxytocin when the gonads were ripe.Then collect the egg and observe the egg color to find the proportion of different eggs in each family and and the genetic regularity of the egg color of the abalone was clarified.It provides theoretical support for the breeding work of the abalones.The results showed that the gonad of the male abalone was mostly white or milky yellow,and the females had more color types: The number of green eggs was the most,the gray eggs and blue eggs were centered,and other colors(such as brownish red,dark gray,beige,etc.)were less.In addition,30 abalones in the family 1 of F1 were selected to determine whether the color of gonad was determined by pigment based on the HPLC,and to analysed the types and content of pigment in the gonad.The results showed that all the individuals of the corrugated abalone contained the fucoxanthin and another unknown pigment.There was a large difference in pigment species between male and female of the wrinkle plate,and the male abalone contained only the cyanobacteria and another unknown pigment,while the female abalone contained other pigments,and there were differences in the pigment types of the female abalone with different egg colors.
