| Inhibin(INH)is a heterodimeric glycoprotein hormone which is secreted by Sertoli cells of male animals and Ovarian Granulosa cells of female animals.Inhibin can specifically act on the pituitary gland to inhibit the synthesis and secretion of follicle-stimulating hormone(FSH)and be in does dependent.Inhibin can also directly act on gonads and influence their functions by paracrine and autocrine.Inhibin immunization can improve the level of FSH by specificity weakening the inhibitory effect of endogenous inhibin and enhance animals reproductive capacity.This paper was to explore the effects of inhibin immunization on associated hormone levels,superovulation and testicular development in Chongming white goat.This paper is divided into the following two parts:1.The effects of inhibin immunization with routine superovulation(CIDR-FSH-PGF2α-LH)on superovulation in Chongming white goatChongming white goats(n=10)were immunized with recombinant inhibin protein twice on d 1 and d 21 and control group(n=10)was set up.CIDR was planted on d 21 and FSH three days diminishing method superovulation started on d 32.Surgical operation and collection of embryos were started on 48 h after mating.During the experiment,the levels of serum FSH,luteotropic hormone(LH),anti-Mullerian hormone(AMH),progesterone(P)and estradiol(E2)were tested.The numbers of ovulation point and recovered embryo of one side ovary were detected when embryo flushing.Results showed : the numbers of one side ovary ovulation point and recovered embryo in immune group were 11.69±1.40 and 9.88±1.64,respectively,which were much significantly higher than those in control group(8.20±1.44 and 6.92±1.62,P<0.01).Serum concentrations of FSH and AMH in immune group were higher than control group(P<0.05),and there were no significant differences in the levels of LH,P and E2 between the two groups(P>0.05).2.The effects of inhibin immunization on reproductive performance using active inhibin immunization on Chongming white male lambsMale lambs(n=6)were immunized with recombinant inhibin protein on d 0,d 21,d 42,d 63 for 4 times and control group(n=6)was set up.During the experiment,body weight(BW),scrotum circumference(SC)and the levels of serum FSH,LH,testosterone(T)were tested and the paraffin sections of testicular tissue were making after castrating on d 84.The expression of reproductive gene in testicular tissue was also tested.Results showed : there were no significant differences on BW and SC between immunized group and control group during the experiment(P>0.05).At castrating,there were no significant differences in BW(21.87±2.19 kg vs 23.58±5.13 kg),SC(20.95±0.74 cm vs 21.57±1.65 cm)and testes weight(TW)(56.88±4.96 g vs 62.99±4.17 g)between immunized group and control group(P>0.05).The paraffin sections of testicular tissue showed that there were no significant differences in histology between the two groups.In serum hormone levels,FSH and T in immunized group were much higher than control group on day 7 after second immunization(P<0.05),however there were no significant differences on other time points.In serum LH level,there were also no significant differences between the two groups.The relative expression levels of Dmrtc2 and SOX30 genes in immunized group were up-regulated(3.04 vs 1.00,P<0.01 and 1.48 vs 1.00,P<0.05)and the relative expression levels of AR gene was down-regulated(0.59 vs 1.00,P<0.05),and there were no differences in the relative expression levels of FSHR,LHR,Gpx4 and DDX4 genes between immunized group and control group(P>0.05).Conclusion : Inhibin immunization could improve the serum levels of FSH and AMH,and then increased superovulation response in Chongming white goat.There was no promotion on the reproductive performance of Chongming white male lambs after inhibin immunization. |
