Ghrelin is a hormone that is first found in the stomach and plays a functional role in combination with growth hormone secretagogue receptor(GHS-R).A lot of research has shown that Ghrelin has many biological effects,such as regulating growth hormone(GH),stimulating appetite and regulating energy balance,affecting insulin secretion,regulating glucose metabolism,anti-inflammatory immunity,promoting heart function and protecting digestive tract mucosa.Red deer is a kind of precious ruminant,and there are few reports on Ghrelin of red deer.To detect the full-length sequence of red deer Ghrelin cDNA and the possible expression of Ghrelin in the body,this study was carried out by cloning,RT-PCR,RT-qPCR and bioinformatics analysis.The results showed that the full-length cDNA sequence of red deer Ghrelin was obtained for the first time.It was a total of 539 bp and contained a 46 bp 5?untranslated region(5?UTR),a 351 bp open reading frame(ORF)and a 128 bp 3?UTR.The ORF encodes 116 amino acid residues,including a signal peptides of 23 amino acid residues,a mature peptides of 27 amino acid residues and a C-terminal peptides of 66 amino acid residues.Bioinformatics analysis shows that there are transmembrane regions and signal peptides in the gene coding products of red deer Ghrelin.RT-PCR and RT-qPCR results showed that Ghrelin mRNA was all expressed in tongue,esophagus,rumen,reticulum,omasum,abomasum,duodenum,jejunum,ileum,cecum,colon,rectum,the brain,hypothalamus,trachea mucosa,lung,myocardial,endocardium,liver,pancreas,spleen,kidney,ureter,bladder,fallopian tube,uterus,vagina,hypophysis,pineal body,submandibular gland,parotid gland,thyroid gland,lymph node,adrenal gland,ovary and skeletal muscle of red deer.The expression level of Ghrelin in abomasum was sifnificantly higher than in other organs.The above results provide a basis for further exploring the related bioinformatics of Ghrelin gene,and provide an effective basis for studying the physiological functions of Ghrelin in the animal body.
