Prunus tenella Batsch.Also known as Xinjiang wild almond.It is a unique wild plant facing extinction in China’s Xinjiang region.Its origin is very old.It is one of the rare plants in the world.Prunus tenella Batsch.(wild Almond)belong to Rosaceae,Prunoideae,Amygdalus,A.ledebouriana Schlecht With the continuous deterioration of human and natural ecological environment,there is an imminent danger of extinction.Therefore,how to adopt scientific protection and scientific methods,which is very important to protect wild almond in Xinjiang.The experimental materials of this study came from three individual colonies in Xinjiang,including Tower City,Tori and Haba River.Through homologous cloning technology,a group of wild almond was used as experimental material to clone its self-incompatibility and related gene Cullin1,and bioinformatics software was used for analysis.The above experiments have laid a certain theoretical foundation for further deepening the scientific protection of wild almond in Xinjiang and exploring its mechanism of self-incompatibility.Pollen of wild almond was used as experimental material in this experiment.First the quality of its pollen total RNA extraction and filtering,then retroviruses to c DNA,and to obtain its conservative middle segment by RACE technique joining together the length of the experiment aim gene c DNA sequence,and then use the NCBI comparison analysis to predict gene of start and stop codon position.Finally,a new PetCullin1 gene c DNA was successfully cloned from wild almond in Xinjiang.Acquired by Xinjiang wild almond Cullin1 gene bioinformatics analysis,the length of the gene size of2677 bp,the biggest open reading frame of 2217 bp,G+C content is 39.9%,A+T content is 60.1%,coded738 amino acids.The molecular formula of PetCullin1 protein is C3887H6064N1028O1133S34.The total number of atoms and relative molecular weight of this protein are 11095 and 20538 respectively,and the number of positively charged residues is 113.The number of residuals with negative charge is 108,while the half-life of its theoretical derivation in rational analysis is 30 h.PetCullin1 protein belongs to water-soluble protein,its hydrophobic parameters for 0.452,fat and unstable parameters were 88.10,40.19,no signal peptide and excluding across a membrane structure,its phosphorylation sites are mainly composed of serine and threonine and tyrosine is complementary.PetCullin1 protein is mainly found in the cytoplasm,and a small amount in mitochondria.The secondary structure of PetCullin1 protein was 60.98%and 6.64%respectively,The irregular curl was 22.36%and the extension chain was 10.03%,Through the above of Xinjiang wild almond PetCullin1 gene bioinformatics analysis,for the types of Xinjiang wild almond’s research,as well as a more scientific protection corresponding foundation for endangered species.
