The study was conducted to investigate the effects of FGF5 and MSTN knockout on the adaptability,growth performance and blood biochemical indexes of cashmere goats,and provided theoretical evidences for exploring whether gene editing has an impact on animal production.Experiment 1: Determination and Study of Adaptability Indexes of of FGF5、MSTN Gene Knockout in Cashmere GoatsIn this study,8 Rams from cashmere goats(hereinafter referred to as positive sheep)and 8 ewes were knockout of the FGF5、MSTN gene;5 Rams were not found in Cashmere goats(hereinafter referred to as negative sheep)Five ewes were selected as study objects,and were selected at-20 ~ ℃-15 ℃,-15 ~ ℃-10 ℃,-10 ~ 0 ℃ ℃,0 ~ 10 ℃ ℃,10 ~ 15 ℃ ℃,15 ~ 20 ℃ ℃ temperature,the above three types of test sheep body temperature,respiratory rate,pulse was strictly measured.Followed by 10 consecutive days of the above three types of test sheep feed intake determination,the first three days for the pre-feeding period,seven days after the test period.The results show :The body temperature of positive ram(39.16 ± 0.42 ℃)was significantly higher than that of negative ram(38.93 ± 0.42 ℃)at 0 ~ 10 ℃(P <0.05)± 0.27 ℃)was significantly higher than that of negative ewes(39.04 ± 0.35 ℃)P <0.05,but all were within the reference values.The results showed that the physiological values of cashmere goats and cashmere goats which were gene knocked-in and genetically introduced were not significantly different from each other,but their feed intake was significantly higher than that of cashmere goats.Goats have faster growth and metabolic capacity.Experiment 2: Study on the Growth Performance of of FGF5、MSTN Gene Knockout Cashmere GoatIn this study,positive ewes were tested in group 1(8 positive rams)and negative ones were control 1(9 negative rams);positive ram was test 2(8 positive ewes),negative Sheep was control group 2(7 negative ewes)as the research object.Test group 1 positive ram and control group 1 negative ram from birth every 345 days for 345 days for weight determination.Test 2 groups of positive ewes and control Body weight was measured in two groups of negative ewes 345 days from birth every 20 days.December 15,2015 All lambs(34)Perform measurements of body size and weight,length of hair.The difference between positive and negative sheep weight,body size index and cashmere length correlation analysis.The results show :The 8-month-old positive Cashmere goat that knocked out the FGF5、MSTN gene has the characteristics of fasting and fasting than the negative ones.There was significant difference in body weight between the experimental group and the control group(P <0.05).There is no positive correlation between velvet length and body weight,body size index,but also the sample size is not enough,the correlation is not shown.However,there was a significant correlation between weight and body size of positive and negative sheep.Experiment 3: Study on the Blood Physiological and Biochemical Indexes of FGF5、MSTN Gene Knockout in Cashmere GoatIn this experiment,the positive Cashmere goats(8 male and 8 female goats,experimental group)knocked out the FGF5、MSTN gene and the common cashmere goats(5 males and 5 females,the control group)The objects of the study were 8 blood routine indexes at 10 months old,1 year old and 1.5 years old respectively,and 14 blood biochemical indexes were measured at the same time.The effect of FGF5、MSTN gene knockout on the blood physiological and biochemical indexes of cashmere goats was analyzed.The results show:In the blood routine test,the HCT of positive ram was significantly lower than that of negative ram at 10 months(P <0.05),and the RDW of positive ram was significantly higher than that of negative ram(P <0.05).At 1 year old,positive ram(P <0.05).The MCH of positive ram was significantly lower than that of negative ram(P <0.05).The positive rate of HGB and HCT of positive ram was significantly higher than that of negative ram Sheep(P <0.05).At 1.5 years of age,the WBC of the positive ewes was significantly higher than that of the negative ones(P <0.05).Blood biochemical indicators,but the values were within the normal range,the remaining indicators were not significantly different(P> 0.05);blood biochemical indicators,10 months of age,the positive ewes globulin was significantly lower than the negative ewes(P < 0.05).At 1 year of age,albumin of positive ewes was significantly higher than that of negative ones(P <0.05);at 1.5 years of age,albumin of positive ewes was significantly higher than that of negative ones(P <0.05)Lower than the negative ewes(P <0.05).But the values were within the normal range,the other indicators were not significantly different(P> 0.05).The protein metabolism,sugar,lipid metabolism,liver,gallbladder function and calcium and phosphorus metabolism of cashmere goat knockout FGF5、MSTN knockout gene had no significant effect.There was no significant difference between the positive and negative controls in the physiological and biochemical indexes of the test sheep,indicating the FGF5、MSTN knockout gene by CRISPR / Cas9 technology did not affect the blood physiological and biochemical indexes of cashmere goats.Conclusion: The blood physiological and biochemical indexes of the test sheep are different between males and females at different ages,but the values are within the range of normal fluctuation.There is no significant difference between the positive and negative controls.This indicated that knockout of FGF5、MSTN gene did not affect the blood physiological and biochemical indexes of cashmere goats.Test Conclusions:No significant differences of FGF5、MSTN knockout on the physiological value of cashmere goats were observed that showed FGF5、MSTN knockout had no impact on adaptability of cashmere goats.While the feed intake of positive goats was significantly higher than that of negative goats,indicated that FGF5、MSTN knockout cashmere goats increase the metabolic ability.There was significant difference of body weight between the experimental group and the control group(P<0.05),indicated that FGF5、MSTN knockout has significant impact on metabolic ability and energy conversion of cashmere goats.There was no positive correlation between velvet length and body weight,body size indexes.However,there was a significant correlation between weight and body size of positive and negative goats.There was no significant difference of FGF5、MSTN knockout on the physiological and biochemical indexes of cashmere goats.However,there are significant differences in blood physiological and biochemical indicators to be further studied.
