Desmodii styracifolii Herba,originating from dried arerial dried parts of Desmodium styracifolium in Family Leguminosae,has the effect of eliminating jaundice and removing dampness due to its cool,sweet and tasteless trait.The great effect of removing urinary calculus makes D.styracifolium Herba preferred by the market,which results in its stable increasing consumption.As the wild resources cannot meet the demands of medical treatment and health protection,the herbs in artificial cultivation has becoming the main source of D.styracifolium Herba.However,the actual market demands still cannot be meted as there are some obvious problems with the uneven artificial-cultivation herbs like the pool seed quality and imperfect cultivation techniques.In response to the above phenomenon,our task groups has carried out the research on seed quality,chemical composition,medicinal material production,medicinal quality and so on,aiming to perfect the quality standards and provide technical support for the production process of D.styracifolium Herba.This research emphasizes on the effective quality inspection methods of seed and germchit,cutting nursery method,technique for planting density and fertilization pattern and the proper harvest time and drying.The main findings are summarized as follows:1.The test established quality test and classification standards of the seed of D.styracifolium.Through analyzing 20 batches selected seeds from different habitat,which includes the determination of 1000-grain weight,moisture content,viability and germination percentage,combining with K-Median analysis,the seeds can be divided into 3 levels.The form of primary seeds is germination percentage ? 70%,viability ? 80% and 1000-grain weight?1.70 g.The form for secondary seed is germination percentage between 55-70%,activity between 60-80% and 1000-grain weight ? 1.70 g.The form for third seed is percentage germination between 40-55%,vigor between 45-60% and 1000-grain weight between 1.60-1.70 g.2.The test established quality test and classification standards of the germchit of D.styracifolium.According to seven indicators like plant height,ground diameter,length of main root,number of lateral root and so on,the germchit can be divided into 3 grades.The form of primary germchit is plant height ? 9.0 cm,ground diameter ? 1.7 mm and the main root length ? 10.0 cm.The form of secondary germchit is plant height between 7.0-9.0 cm,ground diameter 1.4-1.7 mm and main root length 9.0-10.0 cm.The form of tertiary germchit is plant height 5.5-7.0 cm,ground diameter 1.0-1.4 mm and main root length 8.0-9.0 cm.3.According to the study of cutting propagation,planting density and fertilization pattern,the test established a key technical system for the regulated production of D.styracifolium.(1)By conducting the contrast tests of similar cutting under different lignification level,chemical reagent and cutting time,as well as determining and analyzing the survival rate of cuttings,we came to the result that lower lignified cutting possess higher survival rate than the higher lignified one.The cuttingwas higher than others.The cutting exposed to 500 mg/L rooting powder reached a higher survival rate of 81.33%,compared with any other.Besides,April is the best time for cutting nursery.(2)The study of planting density set up three experimental groups,whose planting density is 25 cm×30 cm,35 cm×40 cm and 45 cm×50 cm respectively.After some time of cultivation,we determined the yield of D.styracifolium,including plant height,ground diameter,Stem/leaf ratio and dry weight,content of total flavones,total polysaccharide and schaftoside,which showed that per mu yield of the 25 cm × 30 cm group reached 1083.31 kg,total flavones reached 12.94 kg and total polysaccharide reached 3.01 kg.Except for the content of schaftoside and total polysaccharides,25 cm × 30 cm group were significantly higher than other experimental groups.It was concluded that the planting density can significantly affedt the growth of D.styracifolium,and the optimal planting density of D.styracifolium is 25 cm × 30 cm.(3)The fertilizer application study designed different fertilize times of compound fertilizer and carbamide,then we measured and analyzed the biomass and chemical composition of D.styracifolium herba,which showed that the dry weight of control group(without fertilizer)is 265.07 g/strain,the total polysaccharide is 2161.15 mg/strain,total flavonoids is 2621.23 mg/strain and the schaftoside is 645.67 mg/strain.The yield and the chemical composition of the Control Group were significantly higher than the fertilization group,from which we can see that the fertilization had no significant effect on the accumulation of biomass and content of chemical composition.4.This study determined the optimal harvest time and drying method for D.styracifolium.(1)By collecting 8 samples that from the different every half a month from July to October,and analyzing plant height,the ground diameter,the dry weight and the relative content of chemical composition,we found that plant height,ground diameter presented continuous increase condition as the delay of the harvest time while the dry weight decreased significantly after reaching 102.79 g/strain at the beginning of October.Similarly,the content of total polysaccharide and total flavonoids gradually decreased after reaching 1556.55 mg/strain and 1356.17 mg/strain at the beginning of October.These situations indicated that the overall quality of D.styracifolium decreased after the beginning of October,which is the best harvest time in the Pingyuan County of Meizhou.(2)In the test on drying method,we adopted oven-drying,shade-drying and sun-drying to deal with the fresh herbs respectively,after that we measured their relative content of chemical composition and racorded their external appearance.Among all those herbs the drying ones appeared light green in color accompany with good smell,meanwhile their content of total polysaccharide reached 9.03 mg/g,the total flavonoids 35.9 mg/g and the schaftoside 0.43 mg/g.Oven-drying method assured best quality of the herbs compared with other treatments,which illustrated that the best drying method for D.styracifolium is using oven.
