Dried aerial parts of Desmodium styracifolium(Osbeck.)Merr.,family Leguminosae,are a popular traditional Chinese materia medica and have been used as a medicine for a long history in China.Modern pharmacological studies indicated that it had antilithic,antioxidant and antiinflammatory effects.The mode of resource production of D.styracifolium mainly depends on artificial cultivation due to the shortage of the wild resources.However,as a result of the differences in the geography environment,weather conditions and cultivation technique in different culture regions,and the lack of good germplasm,the yield and quality of this medicinal plant are significant different,which seriously affects the clinical efficacy.In this study,the selection and eveluation of good germplasm and basic pharmacological activity of D.styracifolium were studied,and the main results are as follows:1.The germplasm resources with high yield and high quality were preliminarily selected by selective breeding method,which provided the breeding original materials for the cultivar breeding and the scientific data for controlling the yield and quality of D.styracifolium from the source.This experiment was based on the information of agronomic traits(plant length,leaf area,ground diameter,branch number and plant architecture),yield and chemical components content of individual plant.The quality of the medicinal materials was comprehensively evaluated by principal component analysis method,55 lines with good agronomic traits were preliminarily screened and 47 lines from their offspring with higher quality and higher yield compared with those of the control group were further obtained,which could be used as the key materials for breeding of D.styracifolium.2.Different concentrations of daminozide,6-BA and gibberellin were used to spray on the leaves of D.styracifolium.Then the dry weight and contents of six active constituents(Vicenin-1,Schaftoside,Isoorientin,Isoschaftoside,Vicenin-3 and Isovitexin)were determined for multiple comparison analysis,PCA analysis and hierarchical clustering analysis.The results showed that the contents of Vicenin-1,Schaftoside,Isoschaftoside,Vicenin-3 and isovitexin were significantly increased by spraying with 50 mg/L of 6-BA.Among them,the contents of Schaftoside,Isoschaftoside and Vicenin-3 reached the maximum,but the yield was significantly reduced.The treatment with 2000 mg/L of daminozide could significantly increase the contents of Schaftoside,Isoschaftoside and Isoorientin,and the content of Isoorientin reached the maximum of 0.276 mg/g.applications of 6-benzyladenine at 50 mg/L and daminozide at 2000 mg/L had significant effects on overall quality and yield of D.styracifolium,which suggested that foliar spraying with 2000 mg/L concentrations of daminozide could effectively improve the quality and yield of this medicinal plant.3.To evaluate the quality of germplasm and discuss the suitable cultivation areas of D.styracifolium,25 samples from different areas were collected and a systematic quantitative fingerprint method(SQFM)for combined quantification of five major constituents(Vicenin-1,Schaftoside,Isoschaftoside,Vicenin-3 and Isovitexin)of D.styracifolium was developed in this study.The results showed that the samples from different regions had various contents of flavonoids,the flavonoids contents in the samples collected from Guangdong,Guangxi Guigang and Hainan were slightly higher than those from other regions,and the samples collected in Guangdong province had a good quality consistency.Furthermore,the fingerprintantioxidant efficacy relationship of the different samples was investigated by examining the correlation between the common peaks and the antioxidant effect.Twenty-two common peaks were positively correlated with the antioxidant effect,which provided scientific basis for the quality evaluation of germplasm of D.styracifolium.
