Liquid Culture Method For Rapid Determination Of Cabbage Self Inquiry Method Affinity

Brassica oleracea has a large area of cultivation in China and occupies an important position in the vegetable industry.The hybrid cabbage showed heterosis in hybrid production,the traditional method is to use the self incompatible line preparation,the main cross hybridization rate is less than 100%,the seed production in mixed lines of the phenomenon,it is easy to cause the cuts and the loss of parents.Therefore,breeding and utilization of male sterile cabbage quickly replaced the self incompatible line application.Cabbage male sterile maintainer lines with self compatible lines,it is a time-saving way of breeding;but cabbage breeding lacks an accurate and rapid identification of self compatibility method become an obstacle to improve the breeding efficiency barriers.In this experiment,high affinity,cabbage self incompatibility,self incompatibility,self incompatibility and other 4 kinds of highly inbred lines of different affinities between materials of different liquid culture medium,and the number of stigmas of different culture methods and different culture time effect in vitro effects of different degree of self affinity material and pollen.Determination of affinity results were compared and analyzed the self consistency of conventional method,create a liquid culture method for rapid determination of the affinity of cabbage method.This study draws the following conclusions:(1)4 kinds of different degree of affinity self pollen into 20 mL NLN-14 medium,stigma papilla cells produce the optimum pollen germination of callose hinders the stigma for the number 8.(2)screening suitable cultivation liquid medium and high affinity,self selection and self incompatibility of pollen,the optimum culture medium of pollen grain germination is NLN-14,training is the best way to cultivate oscillation.(3)observation on pollen germination time,the height of self incompatible lines and self compatible lines of pollen,pollen germination,cultivation of the best observation time is 48 h.With the gradual increase of the culture time,the pollen germination rate increased gradually,the pollen tube length increases.(4)determined cabbage self compatibility with liquid,quick and accurate method for the determination of the standard of self incompatible line height: P = 10%;self incompatibility 10%<P = 20%;20%<P = 30% self incompatible lines;self incompatible line height P > 30%;L = 160 px of the pollen tube length of pollen germination condition.Experimental study on cultivation of rapid determination of affinity method of cabbage was obtained in liquid at flowering stage,the selection of fully open flowers in 20mLNLN-14 liquid culture medium adding 8 stigma,a flower pollen culture to shake off the NLN-14 liquid medium,to shake off the dry clean.Then the finger shaped bottle is placed at 25 DEG C,2000 Lx light intensity,illumination time is 12h/d,the table(speed 60r/min)on cultured 48 h removed after observation,pollen tube length is larger than 160 px for pollen germination,the germination standard is: self incompatible line height P = 10%;self incompatibility 10%<P more than 20% self incompatible lines;20% <P = 30%;P > 30% highly self affinity.
