Lentinula edodes polysaccharides have been known to posses various pharmacological activities.Meanwhile,there is an association with structural properties and bio-activities of the polysaccharides at different development stages.This work took L.edodes fruiting bodies at different development stages as the research object,the polysaccharide and protein content of the L.edodes fruiting bodies at different development stages were determined,at the same time,the preparation process of lentinan was studied,then the relationship of the structural properties and bio-activities of crude polysaccharide fractions were discussed.The experimental results are as follows:1.In the process of cultivation,the total sugar and polysaccharide content of the first tide was high,and the third tide was high in protein content.2.Alcohol precipitation process of letinan:Concentration ratio was 4:1(mL:g)and final ethanol concentration was 70%.Decolorization of letinan:pH was 3.50,the concentration of activated carbon was 1%,decolorizing temperature was 20 ?,decolorization time was 100 min,and the rotational speed of the rocking bed was 190r/min.The best process conditions for deproteinized by Sevage method were:Deproteinization by Sevage four times.3.The structural properties of polysaccharides from Lentinula edodes fruiting bodies at different development stages:the yield of P20 was the highest,and the content of polysaccharide and beta glucan was the highest in the P50 component,and reached70.29%and 31.93%in the L3,respectively,but the protein content was the highest in the P20 component,up to 12.83%.The viscosity average molecular weight of different alcohol precipitation components decreases with the increase of alcohol concentration,and the viscosity average molecular weight of P20 components is the largest.4.The results of the structural characteristics of the polysaccharides before and after purification:after purification of the L2P20 component,3 polysaccharides were obtained,in which L2P20A was the main component of polysaccharide and did not contain nucleic acid and protein,which was a beta furan glucose configuration.Scanning electron microscopy showed that the surface structure of L2P20B and L2P20C was similar,showing clastic accumulation,while the L2P20A structure was significantly different from L2P20B and L2P20C,showing a blocky mass with convex and wrinkled.In the study of the Congo red complex,L2P20 and L2P20A contain three helix structures,while L2P20B and L2P20C do not contain three helix structures.The viscosity average molecular weights of L2P20,L2P20A,L2P20B and L2P20C are219.28 KDa,83.35 KDa,44.02 KDa,36.64 KDa,respectively.5.All crude polysaccharide fractions had bio-activities,but polysaccharides fractions of large molecular weight were better than small molecular weight at the bio-activities experiment.These results further showed that there was a relationship between structural properties and bio-activities,the?-glucan may be the major factor in bio-activities,but the molecular weight of?-glucan influence bio-activities intensity.6.The results of antioxidant activity and bacteriostasis of polysaccharides before and after purification:The L2P20 component had better antioxidant effect,but the L2P20A component had better bacteriostasis effect on Staphylococcus aureusthe structural properties and bioactivity of polysaccharides from Lentinula edodes fruiting bodies at different development stages were studied.It was found that during the growth and development,the changes of letinous edodes polysaccharides and other components were significant.There is a great difference in the components of each period.Structural characteristics and biological activity of different fractionation of alcohol precipitation polysaccharides.There is a certain relationship between the structure and activity of polysaccharide,and the structural characteristics and biological activity of the purified letinous edodes polysaccharides are obviously different from those before and after the purification.In the research,the accumulation,structure and biological activity of letinous edodes polysaccharides in different growth stages were studied,which provide a theoretical basis for letinous edodes's cultivation,processing and storage,as well as the preparation and utilization of letinous edodes polysaccharides.
