Five male sheep with weight of 55.6 ± 3.2kg and 2 ?3 years old were randomly divided into five groups: according to 5 × 5 Latin squ are,with the same diet,they were respectively fed 0(control),150,300,450 and 600 mg paraformaldehyde /kg grain(each n=5 for each treatment),rumen fluid was taken at 0,1.5,3,5,7,10 hours after feeding,pH,liquid ammonia nitrogen,volatile fatty acids,formaldehyde and polyethylene glycol concentrations and cellulase,outer cellulase,cellobiase,amylase,protease,pectinase and xylanase activity,and protozoa(Bacteroides,Bacillus,Streptococcus and Streptococcus sp.)were determined,and fungus were counted to study the effect of paraformaldehyde on the rumen microbes and digestion of sheep Enzyme activity.The results show that the feeding of 150?300?450 paraformaldehyde and 600 mg /kg diet,free intake of dry matter production of sheep was increased by2.3(P < 0.05),22.1(P < 0.01),7.5(P < 0.01)and-6.6%(P < 0.01);the number of the rumen protozoa was reduced by 14.2,62.5,72.6 and 85.3%(more than all P < 0.01);and the total number of bacteria increased 35.8,93.9,-6.8 and-24.2%(all P < 0.01);copy number of fungi was increased by 2(P > 0.05),5.7(P > 0.05),13.8(P < 0.01)and 26.8%(P < 0.01);ammonia N was decreased by 9.0,12.0,14.9 and 20.9%(all P <0.01);volatile fatty acids was increased by 2.4(P > 0.05),-0.3(P > 0.05),-1.3(P >0.05)and-10.3%(P < 0.01);the activity of endocellulase was increased by 6.3,33.7,-3.9 and-2.0%(all P < 0.01);and the activity of exocellulase was increased by 3.4(P > 0.05),17.5(P < 0.01),-18(P < 0.01)and-25.7%(P < 0.01);the activity of cellobiase was increased by 8.3,23.7,-24.9 and-31.8%(all P < 0.01);the activity of amylase was increased by 2.8,8.6,16.0 and 23.3%(all P < 0.01);the activity of pcetase was decreased 11.8,12.1,11.2 and 11.1%(all P < 0.01);the activity of protease was decreased and 10.9,29.9,49.6 5 4.6%(all P < 0.01).It was concluded from this study that when the dose of paraformaldehyde was added to 300mg/kg,the free intake of sheep was the highest,at which time the number of protozoa decreased and the total number of bacteria was the highest and the cellulase activity was the highest;...
