Objective: Ascites syndrome(AS)is a common nutritional metabolic disease.Intestinal mucosal structural damage leads to increased permeability is the main pathogenic link to promote ascites,so intestinal structure and function are closely related to the occurrence of AS.Compound traditional Chinese medicine can effectively prevent the occurrence of AS.This experiment was to detect the changes of intestinal tight junction proteins and related regulatory factors in AS broilers,and then to study the prevention and treatment mechanism of compound traditional Chinese medicine on broiler ascites syndrome.MethodThe AS model was constructed by the multi-factor method of adding 3% lard and 4% fish meal to basal feed and 0.12% Na Cl to drinking water at the low temperature of 9-11?.A total of 300 1-day-old Ross broilers were adaptive feeding 7 days,then were randomly divided into normal group,AS model group,compound traditional Chinese medicine high and low dose group(fed with AS model group and respectively given a dose of 2,1 m L/kg·bw·d compound traditional Chinese medicine in drinking water)and L-Arg group(fed with AS model group and added 1% L-Arg in diet).At35-day-old,10 chickens were randomly selected from each group,weighed,sacrificed the carotid artery by bloodletting,observed the pathological changes,weighed the heart and calculated the heart ascites index,observed the histopathological changes and detected the morphological indexes in the middle part of the jejunum.The relative expression of NF-?B and tight junction protein(claudin-1,occludin and ZO-1)m RNA in jejunum was detected by real-time PCR.The contents of MMP-9 and TIMP-1 in jejunum were detected by ELISA.Result1.The growth of the broiler in the AS model group were slow,the spirit was weak,the feather were messy,the abdomen was "penguin"-swollen and had a wave feeling,and a large amount of yellow liquid can be extracted from the abdomen.Pathological necropsy showed liver and lung swelling and congestion,right ventricular hypertrophy with pericardial effusion,intestinal tube swelling,intestinal wall congestion or bleeding spots of varying sizes,and the intestines were filled with thin paste contents.The clinical symptoms and pathological changes of broilers in the high-and low-dose groups of compound Chinese medicine and L-Arg group were all relieved to varying degrees.2.Compared with the normal group,the ascites index,jejunal crypt depth,relative expression of NF-?B m RNA and MMP-9 and MMP-9/TIMP-1 protein contents of broilers in the 35-day-old AS model group were significantly increased(P<0.01);body weight and jejunum tissue villus length,velvet ratio,tight junction protein claudin-1,occludin and ZO-1 m RNA relative expression was significantly reduced(P<0.01);compared with AS model group,compound Chinese medicine Both the high and low dose groups and the L-Arg group significantly improved the above indicators(P<0.01 or P<0.05).Conclusion1.During the occurrence of ascites syndrome,the intestinal mucosal structure and function were destructed by the increase of jejunum inflammatory factors in broilers,which leaded intestinal permeability to increase and participates in the development of broiler ascites syndrome.2.L-Arg and compound traditional Chinese medicine could effectively prevent ascites syndrome by reducing bowel inflammatory injury,improving intestinal structure and function and inhibiting intestinal permeability increased.
