As an important food crop and disaster relief crop in China,sweet potato is also a popular food dish,so sweet potato breeding is of great significance.The construction of the genetic map of sweet potato is an important basis for establishing effective molecular breeding technology.The results of this study were as follows:1.A total of 913 pairs of primers with different parents were selected from the initial development of EST-SSR primers of sweet potato 2000 pairs for the analysis of daughter diversity.2.From the PCR polymorphism amplification results of 913 pairs of primers on the offspring,the markers that conform to chi-square statistical analysis and enter the map were selected.A total of 1276 such specific EST-SSR markers were obtained,including 392 paternal markers(separation ratio 1:1),372 maternal markers(separation ratio 1:1)and 512 daughter markers(separation ratio 1:3).3.The F1 separated group of the 27 x puning cuisines of Taiwan was used as the mapping group,and the paternal and maternal linkage group maps were obtained respectively by using Joinmap4.0 mapping software.There were 50 paternal clusters,including 128 EST-SSR,accounting for 32.6% of the total number of markers.There were 2-9 markers on each cluster,with a length range of 0.6-81.6cm,a length of 1649.6cm covering genome,and an average genetic distance between markers of 12.888 cm.There were 31 matriarchal chain groups,including 121 est-ssr,accounting for 32.5% of the total number of markers.There were 2-53 marker sites in each chain group,with a length range of 1.2-192.8cm,the total length of covered genome was 1075.5cm,and the average genetic distance between the markers was 8.888 cm.The innovation of the results of this study: all the commonly used EST-SSR markers were used for mapping;And the obtained linkage map is the one with the highest number of EST-SSR markers,which is 1276.It is also the graph with the longest distance using EST-SSR marker of sweet potato,which is 1649.6c M.
