Sheep meat has the characteristics of low fat,low cholesterol,high protein,unique flavor,easy to digest and deeply favored by consumers.At present,most researchers still use physicochemical properties to make measurements,while few studies use high-throughput sequencing technologies.In this study,the high-throughput transcriptome was sequenced from the longissimus dorsi muscle of the sheep,and the transcriptome of the longissimus dorsi muscle was analyzed by RNA-Seq technology to screen differentially expressed genes among the breeds.Through the GO and KEGG functional enrichment analysis of differentially expressed genes,the signal pathways and biological processes related to meat quality traits were screened out,and then the expression genes related to meat quality traits were screened,Meanwhile,it provided the data theory basis for further studying the gene and function of sheep meat quality.(1)In this study,four different species of sheep's longissimus dorsi muscle were constructed for high-throughput sequencing.Three samples of each species has a total of 220 G of Clean Reads.(2)A total of 2477 differential genes were obtained from the comparison of the longissimus dorsi muscles of sheep,including 47 co-expressed genes.Among them,614 genes were differentially expressed between BM and STH;1395 genes between BM and SNT;619 genes between BM and DBH;812 genes between STH and SNT;540 genes between STH and DBH;and 433 genes between SNT and DBH.(3)Up-regulate and Down-regulate differentially expressed genes for analysis.There are A total of 1079 differentially up-regulated expressed genes and 1581 down-regulated genes.Using BM as a control group,201 genes were up-regulated and 413 genes were down-regulated between BM and STH;390 genes were up-regulated and 1005 genes were down-regulated between SNTs;318 genes were up-regulated and 301 genes were down-regulated between DBH and BW.STH was used as a control group.Between STH and SNT,194 genes were up-regulated and 618 genes were down-regulated.283 genes were up-regulated and 257 genes were down-regulated.While,SNT was used a control group,Between SNT and DBH,225 genes were up-regulated and 208 genes were down-regulated.(4)GO and KEGG enrichment analysis of differentially expressed genes among the cultivars.The results of GO enrichment analysis showed that the genes of differentially expressed genes mainly concentrate on the development of muscle cell tissues and organs,the contraction of striated muscles,carbohydrate metabolism and the process of phosphate metabolism.KEGG Pathway enrichment analysis yielded 61 significant signaling pathways.KEGG enrichment analysis results showed that differentially expressed genes were involved in the regulation of actin cytoskeleton,regulation of pluripotent stem cells,AMPK,chemokines,PI3K-Akt,FoxO and other signaling pathways.(5)The The co-expression network analysis of differentially expressed genes was carried out,and three modules related to meat quality were screened out.The analysis found that the genes of the three modules were highly correlated with meat quality.Do three modules of network analysis,respectively for 83 key genes,through the literature,found 39 key genes in meat quality was reported in the literature have been reported,there is no report of 44 other genes associated with fat,also can be used as the next step research candidate genes related to meat quality traits.
