In this experiment,the tissue culture seedlings of Beilu blueberry and Lanfeng blueberry were used as test materials.Through different hormone treatments,the optimum formulations for callus induction,adventitious bud induction,elongation induction and rooting induction of two blueberries were selected,and the rapid propagation system for tissue culture of two blueberries was established.At the same time,different concentrations of colchicine were used as induction reagent,combined with tissue culture technology to explore the polyploidization induction of blueberry,The treated plants were observed and identified.The main results were as follows:1 The plant height of the Beilu blueberry tissue culture seedlings is higher and the growth rate is relatively faster.The leaves of Lanfeng Blueberry Tissue Culture Seedlings are rounder and larger than the leaves of Beilu Blueberry.2 The effect of mixed hormone on callus induction was better than that of single hormone.The optimum hormone ratio for callus induction in the experiment was: 1.5mg/L 6-BA+2mg/L 2,4-D.3 The best formula for adventitious bud induction was 1 mg/LZT+0.02 mg/LNAA,and 1/2 WPM was chosen as the basic medium.4 When induced elongation,1mg/L ZT can induce higher and more robust tissue culture seedlings.The concentration of 2T/L and 3mg/L ZT induced plant branching increased significantly,and growth was relatively inhibited.5 Rooting induction of blueberry is suitable for low salt concentration,and the best medium for root induction is 1/4WPM+0.5mg/LIBA.6 The tissue culture seedlings treated with colchicine were generally better than the tissue culture seedlings without colchicine treatment.The plant height was significantly increased,the growth rate was faster,the stems were thicker,the number of leaves increased,soluble sugar and protein.And a significant increase in chlorophyll.7 Taken together,the overall growth of plants treated by culture method in different treatment methods is better than other treatment methods.
