Molecular biological identification of Hyalomma was carried out in parts of Xinjiang to identify the species and pathogens.Parasitic ticks on the surface of livestock were collected from 21 collection points in parts of Xinjiang.Firstly,the tick was classified and identified according to morphological methods.Then,the 12 S rDNA and16 S rDNA genes of tick were amplified by PCR.For the confirmed species of Rickettsia,OmpA gene,gltA gene,17 kDa gene and sca1 gene of Rickettsia were detected by PCR.The 16 S rDNA gene and msp4 gene of Anaplasma;The 18 S rRNA gene of Babesia was tested for pathogens,and the positive products were sequenced to determine the species of tick borne pathogens.The three species of Hyalomma were collected in 21 collection points in Xinjiang,including the 6collection points(number: 1,6,8,12,13 and 15)are Hyalomma detritum;The 6 collection points(number:7,17,18,19,20 and 21)are Hyalomma asiaticum;The 9 collection points(number: 2,3,4,5,9,10,11,14 and 16)are Hyalomma anatolicum.Rickettsia,Anaplasma and Babesia were the pathogenic agents of tick.Rickettsia gltA gene,the test results show that the number for Hyalomma-3 of the Hyalomma anatolicum carried the pathogen with Rickettsia massiliae gathered for a branch,The detection results of the Rickettsia sca1 gene showed that H.detritum,which was number Hyalomma-6,carried pathogens together with Rickettsia raoultii in one branch,The detection results of the Rickettsia OmpA gene showed that Hyalomma anatolicum numbered for Hyalomma-3 and Hyalomma-9 and Hyalomma detritum numbered for Hyalomma-15 carried the pathogen were clustered together with Candidatus Rickettsia barbariae,The detection results of the 17 kDa gene of Rickettsia revealed that H.detritum,which was number Hyalomma-1 and Hyalomma-15,and H.anatolicum,which was number Hyalomma-3,Hyalomma-9 and Hyalomma-10,carried the pathogen together with Rickettsia sp.Hyalomma detritum,which was number Hyalomma-8,carries the pathogen with the Uncultured Rickettsia sp.The results of Anaplasma 16 S rDNA gene test showed that H.anatolicum,which was numbered Hyalomma-5,carried the pathogen together with Anaplasma phagocytophilum,and the results of Anaplasma msp4 gene test showed that H.detritum,which was number Hyalomma-6,carried the pathogen together with Anaplasma ovis.The results of Babesia 18 S rRNA gene test showed that H.asiaticum,which was number Hyalomma-18 was identified as a carrier of Babesia sp.The Hyalomma collected in this experiment,Hyalomma anatolicum were the dominant species in the genus Hyalomma.By combining the morphological characteristics of Hyalomma with the identification results of 12 S rDNA and 16 S rDNA,the ticks could be accurately classified and identified,providing a scientific basis for the composition and distribution of ticks in Xinjiang.Pathogen detection results show that the local Hyalomma carrying Rickettsia massiliae,Rickettsia raoultii,Candidatus Rickettsia barbariae,Rickettsia sp.,Anaplasma phagocytophilum,Anaplasma ovis and Babesia sp.pathogen,suggesting that the parts of Xinjiang region has the popular kinds tick-borne pathogens risk,should according to the local actual situation based on the popular features of ticks media agent,the Hyalomma and pathogen carried of Hyalomma by effective control and prevention.
