| Bovine mastitis is a common and frequently-occurring disease in cattle farm.It can not only lead to the decline of milk yield and quality,but also increase the cost of treatment,elimination rate and mortality,and cause huge economic losses to dairy cow breeding industry.Ru Huangxiao Oral Liquid is a compound Chinese medicine preparation specially developed by the Dairy Cow Disease Innovation Team of Lanzhou Institute of Animal Husbandry and Veterinary Pharmaceutical Research.In order to study its clinical efficacy and pharmacodynamics,acute toxicity,subchronic toxicity,macrophage phagocytosis function,in vitro antimicrobial activity,detumescence,analgesia and target animal test were carried out.1.Acute toxicity test methods include half lethal dose(LD50)method,maximum dose method and maximum tolerance dose method.Results LD50>32000 mg/kg;The maximum tolerance of mice was 150000 mg/kg,which was 75 times higher than that of cows in clinical use.2.Subchronic toxicity test showed that after 30 days of continuous administration of Ru Huangxiao Oral Liquid,there were no significant differences in body mass,organ index,blood physiological index,biochemical index between each group and the control group(P>0.05),and there was no significant change in histopathological observation between the middle and low dose groups compared with the control group,which indicated that Ru Huangxiao Oral Liquid and low dose groups had no significant effect on the growth and development of SD rats.There were no adverse effects on physiological and biochemical indexes and organ index.3.The effects of Ru Huangxiao oral liquid on auricle swelling in mice induced by xylene,cotton ball granuloma,hot plate analgesia,capillary permeability test induced by acetic acid and torsion test induced by acetic acid were studied.The results showed that Ru Huangxiao oral liquid could inhibit acute inflammation,chronic inflammation,central pain and peripheral pain,improve the enhance non-specific immune function and inhibit/kill the main pathogens of cow mastitis.4.In the safety test of target animals,healthy dairy cows were divided into control group,1,3 and 5 times dose groups,which were consistent with the way and days of treatment.The clinical indexes,blood routine and biochemical indexes were observed before and after administration.The results showed that there were no significant differences in the health status and hematological parameters between the different doses of Ru Huangxiao oral liquid and the healthy group(P>0.05),which indicated that Ru Huangxiao oral liquid was safe to target animals at least three times the recommended oral dose.5.The results of clinical treatment showed that the cure rates of Ru Huangxiao oral liquid high,medium and low dose group were 70%,90% and 60%,respectively,and positive drug were 50%;The milk somatic cell count test showed that the number of milk somatic cells in cured dairy cows decreased significantly compared with that before treatment,but it could not decrease to about 200,000/mL in 3-5 days,and the number of milk somatic cells in all high,medium and low dose groups decreased to 200,000/mL in 30 days after curing. |
