| Strawberry(Fragaria ananassa)is perennial herbaceous plants of Rosaceae Fragaria.It has rich nutrition,juicy and fleshy,and it is favored by all over the world,it has a high economic and nutritional value,and also has been planted in large areas in China.The traditional way of self-propagation may lead to the accumulation of strawberry virus,and it causes the decline of strawberry quality and yield,even it also causes the economic losses of farmers.So the Rapid propagation of Strawberry Tissue Culture combined with cryotherapy treatment is an effective way to solve these problems.In this experiment,"Snow White"strawberry varieties were used as experimental materials to establish a tissue culture and rapid propagation system and cryogenic temperature treatment system suitable for local growth of"Snow White"in Shijiazhuang.1.Rapid Propagation System of“Snow White”Strawberry Tissue Culture:The established procedures of the standardization of tissue culture system are as follows:The tender shoot tips which were not invaded by pests and diseases were sterilized.They were cultured on MS+1.0 mg/L BA+0.01 mg/L NAA+6 g/L agar startup medium for 25 days.The shoots were cultured on MS+0.1 mg/L BA+0.01 mg/L NAA+6 g/L agar differentiation medium for 25 days.Then it was trained two generation,and it was incubated on MS+0.1mg/L BA+0.1 mg/L IBA+6 g/L agar rooting medium for 25 days.Then,it was transplanted into a mixture of vermiculite and pastoral soil in equal proportion,and moisturized for 10days.2.Cryotherapy Treatment System of"Snow White"Strawberry:Dark incubation for 3days at 4℃on containing 0.3 mol/L sucrose preculture medium was carried out.60%PVS2was loaded for 1 hour at room temperature,100%PVS2 was dehydrated for 2 hours at 0℃,then it was treated by liquid nitrogen overnight and it was frozen in water bath at 40℃for 2minutes,finally,it was washed three times with liquid MS medium containing 1.2 mol/L sucrose for 10 minutes each time.The highest survival rate was 36.12%.3.The effects of kanamycin and hygromycin on the growth of"Snow White"strawberry seedlings:The effects of kanamycin and hygromycin at different concentrations on the growth of"Snow White"strawberry seedlings were studied.The results showed that the highest resistance concentration of"Snow White"strawberry seedlings on hygromycin was about 150μg/mL,and when it was 150μg/mL,the seedlings began to die and didn’t grow.The highest resistance concentration of"Snow White"strawberry seedlings to kanamycin was about 210μg/mL,and the seedlings were almost albinized at 210μg/mL. |
