Strawberry nutrient-rich, tender and juicy, it is worldwide fruit,has high economic value, in our country has a large area of cultivation.The traditional mode of reproduction is Stolons ramets propagation,the propagation speed is slow, and easy to infection, resulting in reduced yield and quality.By using the technique of Strawberry Tissue Culture Rapid Propagation, can produce a large number of high-quality seedlings in a short period of time, the supply of production.In this experiment, ’Benihoppe’ and ’Shimei7’ strawberry seedlings as test materials,inoculated shoot tip explants,selected the most suitable subculture, subculture interval time and inoculation material types.Studied the hormone combination of the rooting medium and Transplanting conditions. Established the system of tissue culture and rapid propagation of strawberry.By using this system,has produced 5300 tree seedlings. And to calculate the cost of tissue culture, and provide the basis for industrialized seedling.The results of the study show that:1.The early training in subculture,MS as the basic culture medium,adding 6-BA2.0mg/L, IBA0.2mg/L was the most suitable for ’Benihoppe’ strawberry seedlings growth, proliferation multiple was more than 4 times.2. Inoculation ’Benihoppe’ Strawberry petiole with stipules,can produce adventitious bud, but proliferation coefficient significantly lower than the transgenerational seedlings.5.So the optimal inoculation material type is the subculture seedlings per plant.But in order to improve the subculture at the early stage of seedling amount, can be inoculated with stipules.3.In the period of ’Benihoppe’ Strawberry subculture breeding,add 6-BA0.5mg/L, IBA0.15mg/L has the best proliferation effect.The addition of GA3 not only induced stolon, also can improve the proliferation coefficient.GA3 has some losses after high-temperature sterilization,but by increasing the concentration could reach the same effect as filter-sterilized.Considering the convenient operation, can be directly added GA32.0mg/L in the medium(high-temperature sterilization).Sugar concentration on tissue culture medium has effect on the proliferation of tissue culture.Comprehensive cost into consideration, the suitable concentration of granulated sugar is 30g/L.4.The subculture interval of 30 d, 50 d, 60 d had no significant difference in the proliferation coefficient of ’Benihoppe’ Strawberry,but 30 d breeding cycle is short, more conducive to the production.5.’Benihoppe’ strawberry seedlings rooting culture can produce good rooting effect when adding hormone IBA0.16 ~ 0.2 mg/L.The plantlets were appropriate glarecan also contribute to the rooting culture.6.The matrix is vermiculite and peat ratio of 1:1~3:1,the most suitable ’Benihoppe’ strawberry root seedling transplanting,and the survival rate of more than 98%.The transplanting seedlings cultured in light intensity 45000~55000lx,matrix is vermiculite and peat ratio of 1:1~3:1,and irrigated with nutrient solution conditions,the strawberry seedlings grow stronger.7.The reproductive system was established based on experiment, has carried on the ’Benihoppe’ and ’shimei7’ strawberry tissue culture propagation.8.The cost accounting based on the test data, the total cost is about 9633.79 yuan.Transplanting survival of strawberry seedlings of about 5300 trees,and each plant tissue culture seedlings of strawberry production cost is 1.82 yuan.13.The cost of labor costs>water and electricity costs>equipment depreciation costs>medium configured costs> other costs> transplanting costs.
